新城疫弱毒疫苗La Sota株对血清4型禽腺病毒致病作用的影响研究

摘　　要：Ⅰ群血清4型禽腺病毒(FAdV-4)是常见的家禽垂直传播性病毒,感染鸡后可导致心包积液综合征。在生产中,先天感染FAdV-4的雏鸡免疫La Sota疫苗时会明显加重病情,因此我们推测新城疫弱毒疫苗的免疫具有加重或诱发和垂直传播感染的FAdV-4的致病作用。本研究随机选择50枚SPF鸡胚通过卵黄囊接种来模拟FAdV-4先天性感染,同时以接种生理盐水SPF鸡胚作为对照。待孵化出雏后,接毒组和对照组各取20只7日龄SPF鸡免疫新城疫Ⅳ系弱毒疫苗(La Sota株),同时各组其余鸡以相同方式接种等量PBS作为对照。结果显示:FAdV-4感染的雏鸡在免疫La Sota株后更容易发生严重的生长抑制和免疫器官损伤,说明免疫La Sota株能显著增强FAdV-4致病性。本研究结果显示,免疫弱毒疫苗La Sota增强了垂直传播感染的FAdV-4致病性,提示家禽养殖业种源净化的重要性。Fowl adenovirus serotype 4 (FAdV-4) is a common poultry vertical transmission virus,which causes hydropericardium syndrome.When raising chicks carrying congenital FAdV-4 infection,immunization with the La Sota vaccine will significantly aggravate the disease.Therefore,we speculate that the administration of the Newcastle disease Ⅳ attenuated vaccine La Sota can aggravate or induce the pathogenicity of vertical transmission of FAdV-4.In this study,50 SPF chicken embryos were inoculated with FAdV-4 via yolk sac,and other 50 SPF chicken embryos were inoculated with saline as controls.On day 7 post hatching,20 SPF chickens from each of FAdV-4 group and saline group were randomly inoculated with the attenuated vaccine La Sota strain and other chickens in both groups were inoculated with PBS.The results showed that severer growth inhibition and immune organ damages were found in chickens previously infected with FAdV-4 and subsequent immunization with La Sota strain,indicating that administration of La Sota strain significantly aggravated the congenital FAdV-4 infection in chickens.The enhancement of the pathogenicity of FAdV-4 infection by the attenuated immune vaccine La Sota implies the importance of eradication of vertical transmission of FAdV-4.
