摘 要:本研究通过克隆、表达、纯化日本血吸虫胚胎致死异常视觉基因1和2(SjELAV-like 1和SjELAV-like 2),并将获得的纯化蛋白rSjELAV-like 1和rSjELAV-like 2与206佐剂混合免疫BALB/c小鼠,评估rSjELAV-like 1和rSjELAV-like 2的免疫保护效果。结果显示:rSjELAV-like 1和rSjELAV-like 2在减轻宿主虫荷和肝卵方面均有一定的免疫保护作用,rSjELAV-like 2免疫可明显影响虫卵孵化能力;在第1次免疫后rSjELAV-like 2 的特异性IgG抗体水平与对照相比,呈显著升高,具有统计学意义。本研究结果表明,rSjELAV-like 2 比rSjELAV-like 1更适合作为疫苗候选分子,为日本血吸虫疫苗的研发奠定了物质基础。To evaluate the immunoprotection of embryo lethal abnormality visual genes 1 and 2 (SjELAV-like 1 and SjELAV-like 2) of Schistosoma japonicum,these genes were cloned,expressed and the recombinant proteins rSjELAV-like 1 and rSjELAV-like 2 were purified to evaluate their immunoprotection.The purified proteins were emulsified with 206 adjuvant and injected into mice via subcutaneous route.The results showed that both recombinant proteins had a certain effect on reducing worms and eggs burdens.rSjELAV-like 2 immunization effects eggs hatching.Moreover,the immunized mice developed specific IgG antibodies to rSjELAV-like 2 post the first immunization as determined in ELISA.This study evaluated the immunoprotective effects of rSjELAV-like 1 and rSjELAV-like 2,and rSjELAV-like 2 was more suitable as a vaccine candidate for S.japonicum.
