摘 要:为研究不同季节岩羊肠道菌群的多样性,选取同一圈养条件下的5只岩羊作为实验对象,收集新鲜粪便,提取总DNA,PCR扩增后,利用Illumina Miseq测序平台,对16S rRNA基因V3-V4区域序列进行测定。结果显示:在岩羊肠道中检测到的微生物共属于25个门,其中厚壁菌门(Firmicutes)(72.73±5.84)%和拟杆菌门(Bacteroidetes)(16.61±4.37)%为优势菌门;岩羊肠道微生物中含有264个菌属,以Clostridium_XIVa(5.99±2.10)%、Bacteroides(2.62±0.96)%、Clostridium_IV(2.42±1.00)%和Alistipes(1.80±0.78)%为主;通过综合各种不同的多样性指数发现,夏季的肠道菌群丰富度高于其他季节;从不同季节岩羊肠道优势菌群的趋势图可知,夏秋两季岩羊肠道优势菌属变化最大。本研究结果表明,季节变化与岩羊肠道微生物丰富度、多样性及优势菌群组分的变化密切相关。Blue sheep (Pseudois nayaur) is one of the Class Ⅱ national protected species in China and the intestinal microorganisms are essential to the animal health.In this study,fresh feces were collected from 5 blue sheep and the intestinal microorganisms were identified by sequencing 16S rRNA to examine the influence of different seasons on their diversity.Total 25 phyla were detected,among which the Firmicutes and Bacteroidetes were the dominant species with the positive percentage of (72.73±5.84)% and (16.61±4.37)%.Furthermore,264 genera were found in all samples.Clostridium_XIVa,Bacteroides,Clostridium_IV and Alistipes were the major genera accounting for (5.99±2.10)%,(2.62±0.96)%,(2.42±1.00)% and (1.80±0.78)%,respectively.The presence of these intestinal microorganisms was higher in summer than other seasons.The alteration of these genera was more obvious in summer and autumn compared with spring and winter.Collectively,the composition and diversity of the intestinal microorganisms in blue sheep were susceptible to the variation of seasons.
