摘  要:目的检测早期膝骨关节炎大鼠滑膜组织中的S100A12、IL-1β、Galectin-3水平表达,探讨大鼠滑膜组织中三种炎性细胞因子与早期膝骨关节炎相关性的研究。方法 28只成年雄性Wistar大鼠,随机数字表法分成对照组、实验组。实验组大鼠采用左膝关节腔内注射木瓜蛋白酶诱导早期膝骨关节炎;对照组大鼠相同方法及部位注射等量生理盐水。分别于造模成功后第1、28天提取大鼠滑膜组织,采用免疫组化法和酶联免疫吸附法检测滑膜组织中的S100A12、IL-1β、Galectin-3水平表达。结果①实验组造模成功后第1天检测S100A12、IL-1β、Galectin-3水平表达均明显高于对照组,差异有统计学意义(P<0.05);②实验组造模成功后第28天检测S100A12、IL-1β水平表达均高于对照组,差异有统计学意义(P<0.05),但Galectin-3水平表达仍较明显,差异有统计学意义(P<0.05)。结论滑膜组织中S100A12、IL-1β、Galectin-3三种炎性细胞因子均与早期膝骨关节炎密切相关,为临床早期诊断膝骨关节炎提供理论依据,同时明确Galectin-3在早期膝骨关节炎病情进展中的持续表达。Objective To detect the expression of S100A12,IL-1β,Galectin-3 in synovial tissue of early knee osteoarthritis rats,and to explore the correlation between three inflammatory cytokines in synovial tissue of rats and early knee osteoarthritis.Methods 28 male Wistar adult rats were divided into control group (CG) and experimental group (EG) by random number table method.Rats in EG were injected with papain into the left knee joint cavity to induce early knee osteoarthritis,while rats in CG were injected with the same amount of normal saline in the same method and location.Rat synovial tissues were extracted on the 1st and 28th day after successful modeling.Immunohistochemistry and enzyme-linked immunosorbent assay were used to detect the expression of S100 A12,IL-1β and Galectin-3 in the synovial tissues.Results ①The expression levels of S100A12,IL-1β and Galectin-3 in EG were significantly higher than those in CG on the first day after modelingwith statistically significant difference (P<0. 05) .②The expression levels of S100A12 and IL-1β in the experimental group were higher than those in the control group on the 28 th day after successful modelingwith statistically significant difference (P<0. 05),but Galectin-3 level expression was still significantwith statistically significant difference (P<0. 05).Conclusion Three inflammatory cytokines (S100A12,IL-1β and Galectin-3) in synovial tissue are closely related to early knee osteoarthritis,which provides a theoretical basis for the early diagnosis of knee osteoarthritis.Meanwhile,Galectin-3 in early knee sustained expression in the progression of osteoarthritis is clarified.
