摘  要:目的通过研究蒙药蓝刺头对去卵巢大鼠骨密度(bone mineral density,BMD)、血清脱氢表雄酮(dehydroepiandrosterone,DHEA)、肾上腺中芳香化酶P450(aromatase cytochrome P450,CYP450)的作用,探讨蓝刺头防治绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)作用机制。方法取Wistar雌性大鼠56只,随机分为蒙药蓝刺头低、中、高剂量组、假手术组、模型组、己烯雌酚组、强骨胶囊组,共7组,复制去卵巢大鼠骨质疏松症(osteoporosis,OP)模型,行阴道上皮角化实验,确定造模是否成功。确定造模成功后饲养90天,然后灌胃90天后取血,采用酶联免疫法测血清DHEA,处死大鼠分离右侧股骨双能X线测BMD,免疫组织化学法测肾上腺中CYP450表达。结果蒙药蓝刺头提高去卵巢大鼠血清DHEA,增强肾上腺CYP450表达。结论蒙药蓝刺头可提高去卵巢大鼠右侧股骨骨密度、血清DHEA,增强肾上腺CYP450表达,说明蒙药蓝刺头具促进雌激素转化酶、前体物质表达作用,对去卵巢大鼠OP治疗作用明显,通过调节雌激素介导的非经典路径防治PMOP,为深入探讨蒙医"补暖补精—清镇赫依—固骨质壮骨"理论,防治PMOP提供了一定的实验依据。Objective To study the effects of Mongolian medicine on bone mineral density(BMD),dehydroepiandrosterone(DHEA) in serum and aromatase P450(CYP450) of adrenal gland in ovariectomized rats,and to explore the mechanism of prevention and treatment of postmenopausal osteoporosis(PMOP) with echinops latifolius.Methods 56 Wistar female rats were randomly divided into7 group:low,medium and high dose of echinops latifolius group,shame operation group,model group,diethylstilbestril group and Qianggu capsule group.The model of osteoporosis(OP) in ovariectomized rats was established.The keratinization of vaginal epithelium was carried out to determine whether the modeling was successful.After successful molding,the rats were fed for 90 days.And blood was collected after intragastric administration for 90 days.DHEA in serum was measured by enzyme-linked immunoassay.Rats were sacrificed to separate the right femur for measuring BMD by X-ray.The expression of CYP450 in the adrenal gland was detected by immunohistochemical staining.Results The Mongolian medicine echinops latifolius increased the content of DHEA in serum and enhance the expression of CYP450 in ovariectomized rats.Conclusion Mongolian medicine echinops latifolius can increase bone mineral density of right fumer and the content of DHEA in serum,and enhance the expression of CYP450 in ovariectomized rats.It suggests that the Mongolian medicine echinops latifolius has the effect of promoting the expression of estrogenase and precursor substances,having obvious effect on the treatment of OP ovariectomized rats.It prevent and cure PMOP by regulating estrogen-mediated non-classical pathway,providing certain experimental basis for the in-depth study of Mongolian medicine theory of ’tonifying warm and supplementing body essence-clearing and stabilization for Khii-reinforce sclerotin for strengthening bone’and the prevention and treatment of PMOP.
