摘  要:试验选用寒地穗重型超级稻品种松粳9号和穗数型优质品种龙稻18号,通过盆栽试验在灌浆初期喷施外源激素6-苄基腺嘌呤(6-BA)和脱落酸(ABA),齐穗期增施氮肥及喷清水对照处理,系统比较分析外源激素对水稻籽粒淀粉和蛋白质合成相关酶基因及OsRSR1基因转录表达量的影响,旨在阐明外源激素对水稻籽粒碳氮代谢相关酶基因转录表达的影响并为建立优质高产水稻栽培技术提供理论依据。结果表明,水稻籽粒OsGBSS1、OsSSSI、OsISA1、OsAGPL2、OsSBEIIb和OsGS1;3基因mRNA表达量随灌浆进程呈先升后降单峰曲线变化趋势,在灌浆中期表达量最高;转录因子OsRSR1基因mRNA表达量随灌浆进程呈先降后升V字形变化趋势,在灌浆中期表达量最低;喷施6-BA、ABA和增施氮肥均显著上调OsGBSS1、OsSSSI、OsISA1、OsAGPL2和OsSBEIIb基因mRNA表达量,上调幅度达22.35%~525.97%,显著下调转录因子OsRSR1基因mRNA表达量,下调幅度达3.45%~27.93%;喷施6-BA和增施氮肥显著上调籽粒OsGS1;3基因mRNA表达量,上调幅度达241.79%~457.24%,喷施ABA显著下调籽粒OsGS1;3基因mRNA表达量,下调幅度达13.12%~29.55%,基因转录表达量调控是外源激素和氮素营养等对生物性状调控作用的分子基础,外源激素或氮素营养对稻米蒸煮食味品质的影响通过籽粒碳氮代谢相关酶基因转录表达量调控实现。In this pot experiment,the heavy-panicle cultivar Songjing9 with high yield and the multiple-panicle cultivar Longdao18 with good quality in cold region were used to comprehensively investigate the transcription expression levels of genes related to starch and protein synthesis and OsRSR1 under the treatments of 6-benzylaminoprine and abscisic acid at the early filling stage,and the nitrogenous fertilizer treatment at full heading stage,to provide a theoretical basis for elucidating the effects of exogenous hormones on the genes of carbon and nitrogen metabolism in rice grains and for establishing good-quality and high-yield rice cultivation techniques.The results showed that the expressions of OsGBSS1,OsSSSI,OsISA1,OsAGPL2,OsSBEIIb and OsGS1;3 genes in rice grain increased first,then decreased,and reached the peak in the middle filling stage;while the expression of OsRSR1 gene varied as a trend of V symbol,decreased first,then increased,and exhibited the lowest expression level in the middle filling stage;applications of 6-BA,ABA and nitrogen application could significantly up-regulate the expression level of OsGBSS1,OsSSSI,OsISA1,OsAGPL2 and OsSBEIIb gene from 22.35%to 525.97%,and significantly down-regulate the expression level of OsRSR1 gene from 3.45%to 27.93%.Furthermore,the expression level of OsGS1;3 gene was significantly increased to the range of 241.79%and 457.24%by application of 6-BA and nitrogen fertilizers,but significantly decreased to the range of 13.12%and 29.55%under ABA treatment.The regulation of gene transcriptional expression was the molecular basis of the manipulation of biological traits underlying exogenous hormones and nitrogen nutrition,which had an essential effect on rice cooking and eating quality by affecting the transcriptional expression levels of carbon-nitrogen metabolism genes in grains.
