摘  要:本试验旨在研究黄粉虫不同发育阶段脂肪酸合成关键基因的表达,分析脂肪酸合成相关通路关键基因的特性及表达特征。利用Illumina HiSeqTM2000高通量测序平台对不同发育阶段的黄粉虫个体进行转录组测序,并将测序结果与Uniprot以及KEGG数据库进行分析,利用定量PCR检测黄粉虫脂肪酸合成关键基因在不同发育阶段的表达特征。结果显示:本次测序共获得156761个转录子与104937条unigenes。将unigenes与Uniprot数据库进行比对,成功注释了32806条unigenes,并同时鉴定了14条乙酰辅酶A羧化酶(ACC)、75条脂肪酸合成酶(FAS)、101条脂酰辅酶A还原酶(FAR)以及75条脂肪酸去饱和酶(FAD)基因。通过KEGG数据库进行注释,共有44586条unigenes可成功匹配到331条代谢通路中。定量分析发现黄粉虫的ACC与FAS基因在雄成虫期的相对表达量显著高于其他发育阶段(P<0.05),FAD基因在雌成虫期的相对表达量显著高于其他发育阶段(P<0.05),表明ACC、FAS以及FAD基因的表达可能与成虫性成熟或成虫化学通讯等相关;黄粉虫的FAR基因在各个发育阶段都有表达,表明该基因参与了各个发育阶段中脂肪酸的合成。本研究结果表明,黄粉虫脂肪酸合成与发育阶段相关,低龄幼虫代谢强度高,脂肪酸合成相关基因相对表达量高。黄粉虫ACC、FAS、FAR及FAD基因在不同发育阶段表达量具有龄期特异性,揭示这些基因与各龄期的特定生理功能相关。The purpose of this experiment was to study gene expression profile at different development stages of Tenebrio molitor(T.molitor),and to analyze the characteristics and expression features of key genes related to fatty acid synthesis pathways.Illumina HiSeqTM 2000 high-throughput sequencing platform was used to sequence transcriptomes of T.molitor individuals at different developmental stages,and the sequencing results were uploaded to the Uniprot and KEGG databases for analysis.Quantitative PCR was used to detect the expression of key genes related to the fatty acid synthesis of T.molitor at different developmental stages.The results showed that totally 156761 transcripts and 104937 unigenes were obtained,and 32806 unigenes were successfully annotated by compared with Uniprot database,and 14 acetyl-CoA carboxylase(ACC),75 fatty acid synthetase(FAS),101 fatty acyl-CoA reductase(FAR)and 75 fatty acid desaturase(FAD)genes were identified.A total of 44586 unigenes were successfully matched to 331 metabolic pathways through the KEGG database.Quantitative analysis showed that the relative expression levels of ACC and FAS genes in the male adult stage was significantly higher than those in other developmental stages(P<0.05),while the relative expression level of FAD gene in the female adult stage was significantly higher than that in other developmental stages(P<0.05),it showed that the ACC,FAS and FAD genes may be played important roles at adult stage of T.molitor.FAR gene was expressed at all developmental stages,it showed that the FAR gene was involved in fatty acid synthesis in each developmental stage.The results of this study indicate that fatty acid synthesis of T.molitor is related to the developmental stage,the metabolic intensity and the relative expression of fatty acid synthesis-related genes of young larvae are high.The expression levels of ACC,FAS,FAR and FAD genes of T.molitor at different developmental stages are age-specific,revealing that they are related to specific physiological functions of each a
