摘　　要：目的 探讨人输卵管妊娠滋养细胞体外培养的方法与鉴定.方法 取在贵州中医药大学第一附属医院妇科住院部住院的早期输卵管妊娠3例患者的绒毛组织,采用胶原酶-胰酶混合消化法消化细胞,差速贴壁法纯化分离细胞,最后通过免疫荧光共聚焦方法对培养出的细胞进行细胞鉴定.结果 免疫荧光双标记染色检测到角蛋白与波形蛋白共表达.结论 通过显微镜和免疫荧光检测细胞外形和骨架,初步判定该细胞可能为滋养细胞.Objective To explore the methods and identification of tubal pregnancy trophoblasts in vitro.Methods The villi of 3 patients with early tubal pregnancy admitted to the Department of Gynecology,the First Affiliated Hospital of Guizhou University of Chinese Medicine were taken.Collagenase-trypsin mixed digestion method was used to digest cells,and differential adherence method was used to purify and isolate cells.Finally,the cells were identified by immunofluorescence confocal method.Results Co-expression of keratin and vimentin was detected by immunofluorescence double-labeling staining.Conclusion Microscope and immunofluorescence can detect cell shape and skeleton.It is preliminarily determines that the cell may be a trophoblast cell.
