摘  要:目的基于体外实验探讨补阳还五汤对血管新生的影响。方法体外培养鼠胸主动脉段,给予补阳还五汤载药血清干预评估其对血管新生的影响;体外培养人脑微血管内皮细胞(HBMECs),给予补阳还五汤载药血清干预,采用细胞计数8(CCK-8)、划痕迁移法和三维细胞培养法评估补阳还五汤对内皮细胞增殖、迁移和成管的影响;采用反转录-聚合酶链反应(RT-PCR)对血管生成因子进行检测,分析补阳还五汤对促血管新生相关因子表达的影响。结果各组血管段均可观察到新生血管生成,与对照组比较,补阳还五汤干预后存活的新生血管数量明显增多;体外内皮细胞干预结果显示,补阳还五汤可促进HBMECs的增殖、迁移和成管,促进血管内皮生长因子(VEGF)表达。结论补阳还五汤具有促进血管新生的作用,该作用可能与上调VEGF表达有关。Objective To explore the effect of Buyang Huanwu Decoction(BYHWD)on angiogenesis in vitro.Methods The effect of BYHWD on angiogenesis was analyzed by rat thoracic aortic ring assay.The proliferation,migration,and tube formation of human brain microvascular endothelial cells(HBMECs)were performed by cell counting kit-8(CCK-8),scratch-wound migration assay,and three-dimensional cell cultivation,respectively.Furthermore,growth factors related to angiogenesis were analyzed by RT-PCR.Results Neovascularization could be observed in all vessel segment in each experimental groups.Compared with control group,BYHWD could facilitate the formation of neovessels,and promote the proliferation,migration and tube formation of HBMECs.BYHWD could promote the expression of vascular endothelial cell growth factor(VEGF).Conclusion BYHWD can promote angiogenesis which may be related to the up-regulation of VEGF.
