摘 要:目的制备盐酸多西环素缓释微丸并考察其体外释放行为。方法采用挤出滚圆法制备微丸。研究工艺及处方因素对盐酸多西环素的制备和释放度的影响。结果优选最佳工艺参数为挤出速度45 r·min^-1、滚圆速度600 r·min^-1、滚圆时间12 min;优选的处方为MCC 19%、十八醇45%、粘合剂:无水乙醇+EC。试验表明多西环素缓释微丸主药含量和微丸累积释放度均达到预期结果,其释药机制为药物扩散和骨架溶蚀的混合型机制。结论通过以上处方和工艺自制的盐酸多西环素缓释骨架微丸释药平稳、缓慢、持续。OBJECTIVE:To prepare Doxocycline Hydrochloride sustained-release pellets and to investigate its drug release behavior in vitro.METHODS:The pellets were prepared by the extrusion-spheronization method,focusing on the influence of technological procedure and recipe ingredients on the quality of pellets and their release properties.RESULTS:The optimal process parameters were as follows:extrusion velocity of 45 r·min^-1,spheronization velocity of 600 r·min^-1,spheronization time of 12 min.The optimized recipe consists of MCC 19%and stearyl alcohol 45%using anhydrous ethanol+EC as adhesives.Experiment revealed that both the concentration and cumulative percentage release of the main active ingredient of the prepared pellets reach the desired level.The release behavior was identified to be a hybrid diffusion and corrosion mechanism.CONCLUSIONS:Under these preparation conditions,the Doxocycline Hydrochloride pellets allow a stable,slow,and sustained-release.
