摘  要:蛋白质热转移法是一种经济快速简便的确证小分子与靶标蛋白相互作用的实验技术,该技术通过实时荧光定量PCR仪测试小分子和靶标蛋白结合后蛋白质熔解温度的变化,确证活性小分子与靶蛋白质分子间的直接相互作用,为科研常用实验手段,但在本科药物化学生物学实验教学中却较少涉猎。基于教研结合,成果导向教育理念(OBE),设计了蛋白质热转移法的实验教学案例。通过该实验的训练有助于药学专业本科生掌握实时荧光定量PCR仪的使用,巩固利用靶标热稳定性改变确证靶标与药物小分子相互作用的原理,培养学生的药物创新意识、团队协作精神和药学实践能力。Protein Thermal Shift Assay (TSA) is a cheap and convenient method to confirm the interaction between small molecules and target protein.This method can be used to confirm the direct interaction between small molecules and target proteins through measured the protein’s melting temperature change using real time fluorescent quantitative PCR (RT-PCR) machine.TSA is used very often in research work,however,it is seldom involved in undergraduates’ experimental teaching.Based on the direction of teaching-research combination and OBE (outcome based education)conception,a TSA experimental teaching case was designed for medicinal chemical biology experimental teaching.Through running the TSA experiment,the undergraduates majored in pharmacy can master the usage of real-time PCR machine,be familiar with the knowledge of the interaction confirmation between target and small drugs through target’s thermal stability changes,improve their scientific research experience,team work spirit and pharmaceutical practice ability.
