摘 要:目的探讨护心方对心肌梗死后大鼠交感神经重构以及室性心律失常诱发的影响。方法选取45只清洁级SD雄性大鼠为研究对象,将研究对象随机分为假手术组、心肌梗死组、心肌梗死+低剂量护心方组、心肌梗死+中剂量护心方组、心肌梗死+高剂量护心方组。采用对大鼠冠状动脉进行结扎的方法构建心肌梗死模型,术后4周,检测大鼠心肌组织GAP43和TH阳性纤维密度、大鼠心功能、心律失常情况以及大鼠血清中IL-1、IL-6、TNF-α的含量。结果与假手术组相比较,在心肌梗死组的心肌梗死周边区域,GAP43和TH阳性的交感神经纤维分布紊乱,直径粗乱,纤维密度升高;心肌梗死+护心方中、高剂量组心肌细胞交感神经纤维密度较心肌梗死组明显降低。心肌梗死组与心肌梗死+护心方低、中、高剂量组室性心律失常评分明显升高(均P<0.05),而心肌梗死+护心方中、高剂量组室性心律失常评分较心肌梗死组明显降低(P<0.05)。心肌梗死组及心肌梗死+护心方低、中、高剂量组大鼠EF值明显降低(P<0.05);而心肌梗死+护心方低、中、高剂量组大鼠EF值与心肌梗死组无明显差异(均P>0.05)。与假手术组相比较,各组血清IL-1、IL-6及TNF-α水平均明显升高(均P<0.05),而心肌梗死+护心方中、高剂量组血清IL-1、IL-6、TNF-α水平较心肌梗死组明显降低(P<0.05)。结论护心方能减少室性心律失常发生,抑制交感神经重构,其机制可能与抑制心梗后炎症反应有关,而护心方对心肌梗死后大鼠心脏功能无明显影响。Objective To investigate the effects of Huxin Formula on sympathetic nerve remodeling and ventricular arrhythmia induciblity in rats after myocardial infarction.Methods Forty-five male SD rats are selected as the research objects and randomLy divided into sham operation group,myocardial infarction group,myocardial infarction+low dose Huxin Formula group,myocardial infarction+medium dose Huxin Formula group and myocardial infarction+high dose Huxin Formula group.The myocardial infarction model is established by ligating the coronary artery in each group rats.4 weeks after operation,the myocardial tissue GAP43 and TH positive fiber density,the heart function,the ventricular arrhythmia scores,and serum IL-1,IL-6 and TNF-αconcentration are measured.Results Compared with the sham operation group,the GAP43 and TH positive sympathetic nerve fibers at infarct border zones are disorderly distributed and the diameter is coarse in the myocardial infarction group,and the fiber densities are increased.While the sympathetic nerve fiber densities in myocardial infarction+medium and high dose Huxin Formula group are significantly lower than that in the myocardial infarction group.The ventricular arrhythmia scores both in myocardial infarction group and myocardial infarction+low,medium and high dose Huxin Formula groups are significantly increased(both P<0.05),while comparing with the myocardial infarction group,the ventricular arrhythmia scores in myocardial infarction+medium and high dose Huxin Formula groups are significantly lower(P<0.05).EF in myocardial infarction group and myocardial infarction+low,medium and high dose Huxin Formula group significantly decrease(P<0.05).There are no significant differences between in myocardial infarction group and myocardial infarction+different dose Huxin Formula groups(all P>0.05).Compared with the sham operation group,the serum levels of IL-1,IL-6 and TNF-αin each group are significantly increased(all P<0.05),while the serum levels of IL-1,IL-6,TNF-αin medium and high dose Huxin Form
