川陈皮素通过抑制TGF-β1抑制成纤维细胞的生长和迁移

摘　　要：目的探讨川陈皮素对成纤维细胞生长和迁移的调节作用。方法分别使用10、20、40、80μmol/L的川陈皮素作用于成纤维细胞,通过噻唑蓝(mcthyl thiazolyl tetrazolium,MTT)实验检测细胞的增殖情况,Annexin V-PI实验检测细胞的凋亡情况,Transwell实验检测细胞的迁移能力。利用Western blot实验检测川陈皮素对于collagenⅠ、α-SMA和TGF-β1表达的影响。沉默成纤维细胞中TGF-β1后,使用40μmol/L的川陈皮素作用于细胞,采用MTT实验检测细胞的增殖情况和Western blot检测相关蛋白的表达情况。结果与对照组比较,不同浓度的川陈皮素能够显著抑制成纤维细胞的增殖、促进细胞的凋亡并抑制细胞的迁移,其组间比较差异具有统计学意义(P<0.05)。川陈皮素能够抑制collagenⅠ、α-SMA和TGF-β1蛋白水平的表达。沉默成纤维细胞中TGF-β1,使川陈皮素对细胞增殖和相关蛋白的抑制作用减弱。结论川陈皮素能够通过抑制TGF-β1来抑制成纤维细胞的生长、迁移和纤维化。Objective To explore the regulating effect of nobiletin on the growth and migration of fibroblasts.Methods The fibroblasts were treated by nobiletin of 10,20,40 and 80μmol/L,respectively.The proliferation of fibroblasts was detected by MTT assay,the apoptosis was detected by Annexin V-PI assay and the migration was detected by Transwell assay.The effect of nobiletin on the expression of collagenⅠ,α-SMA and TGF-β1 was detectd by Western blot.After TGF-β1 was silenced in fibroblasts,nobiletin of40μmol/L was used to treat the cells.The proliferation of fibroblasts was detected by MTT assay and the expression of related proteins was detected by Western blot.Results Compared with the control group,the proliferation and migration of fibroblasts was significantly inhibited and the apoptosis was significantly promoted by various concentrations of nobiletin.The difference between two groups was statistically significant(P<0.05).The expression of collagenⅠ,α-SMA and TGF-β1 was inhibited by nobiletin.After TGF-β1 was silenced in fibroblasts,the inhibition effect of nobiletin on fibroblasts proliferation and related proteins was decreased.Conclusion The growth,migration and fibrosis of fibroblasts could be inhibited by nobiletin through inhibition of TGF-β1.
