摘 要:铜暴露可损害雄性动物生殖系统。本试验将20只20日龄雄性大鼠随机分为4组,每组5只,分别饲喂含不同铜含量的饲料:对照组(15 mg/kg)、高铜Ⅰ组(30 mg/kg)、高铜Ⅱ组(60 mg/kg)和高铜Ⅲ组(120 mg/kg),连续饲喂6个月,采集大鼠睾丸组织,以探讨长期高铜暴露对大鼠睾丸的影响。进行H&E染色以观察睾丸组织形态学变化,使用qRT-PCR检测睾丸自噬相关基因LC3Ⅰ、LC3Ⅱ、ATG5、Beclin1、p62的表达,使用Western-blot法检测睾丸自噬相关蛋白LC3Ⅱ/LC3Ⅰ、ATG5、Beclin1的表达。试验结果显示,与对照组相比,大鼠体重、睾丸重量及睾丸系数无显著差异;随着饲料铜含量升高,睾丸组织损伤加剧;高铜组自噬相关基因LC3Ⅰ、LC3Ⅱ、Beclin1和p62的mRNA表达水平显著上调(P<0.05),而ATG5 mRNA表达水平呈上调趋势,但无显著差异(P>0.05),自噬相关蛋白LC3Ⅱ/LC3Ⅰ、ATG5、Beclin1的表达水平显著上调(P<0.05)。试验结果表明,长期高铜暴露可诱导雄性大鼠睾丸组织病理学损伤及自噬发生。Copper exposure could damage the reproductive system of male animals.To investigate the effects of long-term high level of copper exposure on male rat testis,twenty male rats aged at20-day-old were randomly divided into 4 groups(n=5),and were continuously fed with different levels of dietary tribasic copper chloride(TBCC)for 6 months:control group(15 mg/kg),high copper groupⅠ(30 mg/kg),high copper groupⅡ(60 mg/kg)and high copper groupⅢ(120 mg/kg),respectively.H&E staining was performed to observe the morphology of testis tissue.The expression in mRNA levels of autophagy-related genes LC3Ⅰ,LC3Ⅱ,ATG5,Beclin1,and p62 were detected by q RT-PCR,and the expression levels of autophagy-related proteins LC3Ⅱ/LC3Ⅰ,ATG5,and Beclin1 were measured by Western-blot.In result,there was no significant difference in body weight,testicular weight and testicular coefficient among the four groups.The damage of testicular tissue was aggravated with the increase of dietary TBCC content.Compared to the control group,the mRNA expression levels of autophagy-related genes LC3Ⅰ,LC3Ⅱ,Beclin1 and p62 were significantly up-regulated(P<0.05),and the expression level of ATG5 tend to be up-regulated,but the difference was not significant(P>0.05).The expression levels of autophagy-related proteins LC3Ⅱ/LC3Ⅰ,ATG5 and Beclin1 were significantly up-regulated when compared with the control group(P<0.05).These results demonstated that long-term high level of dietary TBCC exposure could damage the testicular tissue and induce testicular autophagy in rats.
