摘 要:Hedgehog(HH)信号通路在胚胎发育过程中发挥着重要作用。HH主要有三个配体,其中sonic hedgehog(SHH)主要与肾损伤后修复和肾脏发育相关。近期研究发现SHH信号通路的激活与肾纤维化发生发展有紧密联系。在各种慢性肾脏病(chronic kidney disease,CKD)中,SHH在肾小管上皮细胞中高表达后直接作用于肾间质成纤维细胞。其扮演生长因子的角色并调控各级纤维化相关基因,从而导致细胞外基质沉积。本文将阐述SHH信号通路原理以及在肾脏疾病中的调控。并讨论SHH促进肾纤维化的可能机制以及分析阻断信号通路后的影响。SHH信号通路可能成为CKD患者新的治疗靶点。Hedgehog(HH)signaling pathway plays an important role in embryonic development There are three main ligands in HH,among which sonic hedgehog(SHH)is mainly related to repair and kidney development after kidney injury.Recent studies have found that activation of SHH signaling pathway is closely related to the occurrence and development of renal fibrosis.In various chronic kidney diseases(CKD),SHH is highly expressed in renal tubular epithelial cells and directly acts on renal interstitial fibroblasts.It acts as a growth factor and regulates genes associated with fibrosis at all levels,leading to extracellular matrix deposition This paper will describe the principle of SHH signaling pathway and its regulation in kidney diseases.The possible mechanism of promoting renal fibrosis by SHH and the effect of blocking the signal pathway were discussed.SHH signaling pathway may become a new therapeutic target for CKD patients.
