摘  要:目的:探讨杜仲多糖对2型糖尿病小鼠胰腺组织氧化应激的影响。方法:尾静脉注射低浓度链脲佐菌素(STZ,60 mg/kg),高糖高脂饮食诱导建立2型糖尿病(T2DM)小鼠模型。将成功建立的T2DM小鼠随机分为模型组、二甲双胍组、杜仲多糖高剂量组、杜仲多糖中剂量组、杜仲多糖低剂量组,并以健康昆明小鼠为正常对照组,每组10只。各组小鼠灌胃给药连续28 d,末次给药2 h后检测小鼠空腹血糖(FBG),HE染色及透射电镜观察胰腺组织的病理学和超微结构的变化,试剂盒检测胰腺组织中氧化应激相关指标氧自由基(ROS)、羟自由基的含量及过氧化氢酶(CAT)、超氧化物歧化酶(SOD)的活性。结果:二甲双胍组和杜仲多糖高、中、低剂量组小鼠FBG低于模型组(P<0.01),胰腺组织中CAT、SOD活性高于模型组,而ROS、羟自由基的含量低于模型组(P<0.05或P<0.01)。杜仲多糖高剂量组小鼠SOD的活性高于二甲双胍组(P<0.05),但其FBG、CAT的活性、ROS含量及羟自由基含量与二甲双胍组比较,差异无统计学意义(P>0.05)。HE染色及透射电镜结果显示,与模型组比较,二甲双胍组、杜仲多糖各剂量组的小鼠胰腺组织病理形态学和超微结构改变有所改善。结论:杜仲多糖有降低空腹血糖的作用,其作用机制可能为增加机体的抗氧化因子活性,从而减少氧化应激对胰腺的损伤,进而使胰腺组织损伤得到改善。Objective:To explore the effect of Eucommia ulmoides polysaccharides on oxidative stress in pancreas tissue of type 2 diabetic mice.Methods:Type 2 diabetes mellitus(T2DM)mice model was induced by streptozotocin(STZ,60 mg/kg)tail vein injection and high glucose and high fat diet.The successfully established T2DM mice were randomly divided into model group,metformin group,eucommia polysaccharide high-dose group,eucommia polysaccharide medium-dose group,eucommia polysaccharide low-dose group,and healthy Kunming mice were used as normal control group,10 mice in each group.The mice in each group were administered by intragastric administration for 28 consecutive days,and the fasting blood glucose(FBG)of the mice was detected 2 hours after the last administration.Subsequently,HE staining and transmission electron microscopy were used to observe the pathology and ultrastructural changes of pancreatic tissue.Oxidative stress related indicators radical oxygen species(ROS),hydroxyl radical,catalase(CAT)and superoxide dismutase(SOD)were determined.Results:Metformin group and eucommia polysaccharide high,medium and low dose group mice FBG is lower than the model group(P<0.01),pancreatic tissue CAT and SOD activities are higher than the model group,and the content of ROS and hydroxyl radicals are lower than the model group(P<0.05 or P<0.01).Furthermore,the SOD activity of mice in the high-dose eucommia polysaccharide group was higher than that in the metformin group(P<0.05).However,compared with the metformin group,the FBG,CAT activity,ROS content and hydroxyl radical content of mice in the high-dose eucommia polysaccharide group were not statistically significant(P>0.05).The results of HE staining and transmission electron microscopy showed that compared with the model group,the pathological morphology and ultrastructural changes of the pancreas tissue of the mice in the metformin group and the each dose group of eucommia polysaccharide were improved.Conclusion:Eucommia ulmoides polysaccharides have a certain effect of reduc
