摘  要:目的:基于网络药理学研究北刘寄奴药理作用的物质基础和作用机制。方法:通过中药系统药理学分析平台(TCMSP)筛选北刘寄奴的化学成分、作用靶点、治疗疾病。通过蛋白数据库预测候选活性靶点相互关系,用生物信息学技术分析靶点的功能,通过基因本体(GO)功能和通路富集分析,研究北刘寄奴作用机制。结果:筛选共获得5个有效化合物(其中1个化合物没有相应的靶点),共44个相关靶点,涉及79种疾病。PPI网络中有44个靶点,关键靶点有EGFR、CCND1等。基因功能富集结果分析共涉及12个条目,包括细胞组成5个,分子功能2个,生物过程5个。通路富集分析得到7条通路,包括乙型肝炎、PI3K-Akt信号通路、细胞凋亡等。结论:北刘寄奴中木犀草素等活性成分通过作用于PTGS1、PRSS1、NCOA2等靶点,协同参与多条通路的调节,发挥其抗炎、抗菌等作用。Objective:To study the material basis and mechanism of pharmacological effects of Beiliujinu(Artemisiae anomalae)based on network pharmacology.Methods:TCMSP was used to screen the chemical components,targets and diseases of Beiliujinu(Artemisiae anomalae).The relationship between the candidate active targets was predicted by protein database,the function of the targets was analyzed by bioinformatics technology,and the mechanism of action of Beiliujinu(Artemisiae anomalae)was studied by the analysis of Gene Ontology(go)function and pathway enrichment.Results:A total of 5 effective compounds were screened(one of which has no corresponding target),44 related targets,involving 79 diseases.There are 44 targets in PPI network,and the key targets are EGFR and CCND1,etc.The analysis of gene function enrichment results involved 12 items,including 5 cell compositions,2 molecular functions,and 5 biological processes.Pathway enrichment analysis yielded 7 pathways,including hepatitis B,PI3K-Akt signaling pathway,and apoptosis.Conclusion:The active components of luteolin in Beiliujinu(Artemisiae anomalae),through acting on PTGS1,PRSS1,NCOA2 and other targets,cooperate in the regulation of multiple pathways and exert their anti-inflammatory and antibacterial effects.
