摘  要:目的建立手性HPLC法分离布康唑、抑霉唑、舍他康唑、酮康唑和咪唑乙醇5种手性药物对映对映体。方法使用Chiralcel OJ-H手性固定相,通过考察流动相中极性醇类改性剂种类、烷醇体积比、柱温和流速对分离的影响,优化了分离条件。结果在优化的色谱条件下,5种药物对映体均达到了完全分离,且热力学研究结果表明该5对对映体药物的手性拆分过程均受焓驱动影响,低温有利于其对映体分离。结论Chiralcel OJ-H柱适于分离上述5种手性药物。Objective To establish a high performance liquid chromatography methods for the chiral separation of five imidazole antifungal drugs including butoconazole,imazalil,sertaconazole,ketoconazole and imidazole ethanol.Methods With the application of Chiralcel OJ-H column,the factors affecting separation process such as the type and concentration of organic modifier,column temperature and flow rate were investigated.Results Under the optimal condition,five imidazole antifungal drugs were completely separated.Besides,the thermodynamic study indicated that the enantioseparation of these five drugs was enthalpically driven and showed that low column temperature was beneficial to separation.Conclusion Chiralcel OJ-H column can achieve complete separation of these five imidazole antifungal drugs.
