摘  要:目的研究益气活血方对内毒素(LPS)诱导小鼠炎症及内皮损伤的干预作用。方法50只8~12周龄雄性C57BL/6小鼠随机分为空白对照组、LPS组、益气活血方组[1.29 g/(kg·d)]、吡咯烷二硫代氨基甲酸酯(PDTC,NF-κB抑制剂)组(50 mg/kg)、益气活血方[1.29 g/(kg·d)]+PDTC(50 mg/kg)组,每组10只。益气活血方组及益气活血方+PDTC组中药灌胃2周,其余组等体积生理盐水灌胃,结束后次日PDTC组及益气活血方+PDTC组腹腔注射PDTC,1 h后除空白对照组外其余各组腹腔注射LPS(20 mg/kg)刺激6 h。采集小鼠血清及主动脉,ELISA检测血清炎性因子TNF-α、IL-6浓度,qRT-PCR、Western Blot分别检测黏附因子VCAM-1、ICAM-1及促凝血因子PAI-1、TF的mRNA、蛋白表达水平,并用Western Blot检测NF-κB通路磷酸化水平。结果与空白对照组比较,LPS组血清TNF-α、IL-6浓度,主动脉VCAM-1、ICAM-1、PAI-1、TF mRNA及蛋白表达水平均升高(P<0.01),NF-κB通路基团P65、IκB磷酸化水平亦明显升高(P<0.01);与LPS组比较,益气活血方组、PDTC组及益气活血方+PDTC组血清TNF-α、IL-6浓度,VCAM-1、ICAM-1、PAI-1、TF mRNA及蛋白水平,P65、IκB磷酸化水平均明显降低(P<0.05,P<0.01);益气活血方+PDTC组与益气活血方组比较,差异无统计学意义(P>0.05)。结论益气活血方通过抑制NF-κB通路活性发挥抗LPS诱导小鼠炎症及内皮损伤的作用。Objective To study the effects of Yiqi Huoxue Formula(YQHX)on inflammatory responses and endothelial injury induced by lipopolysaccharide(LPS)in mice.Methods Fifty male C57 BL/6 mice(8-12 weeks old)were randomly assigned to the control group,LPS group,YQHX group(1.29 g·kg^-1·d-^6^1),pyrrolidine dithiocarbamate(PDTC,NF-κB inhibitor)group(50 mg/kg),and YQHX(1.29 g·kg^-1·d^-1)plus PDTC(50 mg/kg)group,10 in each group.The YQHX group and YQHX plus PDTC group were administered with YQHX for 2 weeks by gastrogavage,while the other groups were administered with equal volume of normal saline by gastrogavage.The next day after administration,PDTC group and YQHX plus PDTC group were intraperitoneally injected with PDTC for 1 h.Then,all groups except the control group were intraperitoneally injected with LPS(20 mg/kg)for another 6 h,and serums and aortas were collected,respectively.The levels of TNF-αand IL-6 in the serum were detected using ELISA.The mRNA expression levels of VCAM-1,ICAM-1,PAI-1 and TF were determined using qRT-PCR and the protein levels of these factors as well as the phosphorylation levels of NF-κB pathway were detected using Western Blot.Results The levels of TNF-α,IL-6,VCAM-1,ICAM-1,PAI-1 and TF in LPS group were higher than those in control group(P<0.01),and the phosphorylation levels of P65 and IκB were also increased significantly(P<0.01).Compared with the LPS group,the levels of TNF-α,IL-6,VCAM-1,ICAM-1,PAI-1,TF and NF-κB pathway phosphorylation were decreased in YQHX group,PDTC group and YQHX plus PDTC group(P<0.05,P<0.01).There was no significant difference among groups of PDTC and YQHX plus PDTC(P>0.05).Conclusion YQHX suppresses the inflammatory responses and endothelial injuries induced by LPS in mice by inhibiting the activities of NF-κB pathway.
