摘  要:为了进一步研究OsCAT3的分子机制及分子调控网络,利用基因芯片对WT和OsCAT3crispr幼苗进行了全基因组分析。本研究利用CRISPR/cas9技术敲除过氧化氢酶基因OsCAT3,获得的转基因植株OsCAT3crispr。利用基因芯片技术分析发现,转基因植株OsCAT3crispr中差异表达基因共8 812个,其中4 580个表达上调,4 232个表达下调。对差异表达基因进行GO和KEGG富集分析发现,差异基因在毒素代谢以及光合作用相关的途径中显著富集,表明水稻过氧化氢酶基因OsCAT3可能与毒素代谢和光合作用途径有关。本试验对OsCAT3crispr进行基因芯片分析得到了大量的差异基因,对深入了解过氧化氢酶基因调控机制有重要的意义。For better research the molecular mechanism of OsCAT3 and the molecular regulation mechanism, the whole genome analysis of wild type and transgenic plant seedlings was carried out using gene chip. Using CRISPR/CAS9, we obtained rice OsCAT3 CRISPR/cas knockout plant. The study by using gene chip found that there were8 812 genes with expression differences, of which 4 580 genes were up-regulated and 4 232 were down-regulated.The GO and KEGG enrichment analysis showed that the differentially expressed genes were significantly enriched in biological processes related to toxin metabolism and pathways associated with photosynthesis, which indicated that the rice OsCAT3 may be related to toxin metabolism and photosynthesis pathway. Through the study, the obtained a large number of differential genes is of great significance for further understanding the regulation mechanism of rice catalase gene.
