摘 要:本研究以航天诱变向日葵长花瓣突变体(long petal mutant, lpm)为研究材料,采用抑制性差减杂交(suppression subtractive hybridization, SSH)的方法,成功构建了其幼嫩花芽的SSH文库。从文库中随机挑选399个阳性克隆进行测序拼接获得309个Unique表达序列标签(expression sequence tags, EST)。在GenBank数据库进行相似性比对分析,共有121条EST与已知序列具有较高的相似性,占Unique EST的39.16%。通过对已知基因进行功能分析,推测向日葵花瓣的形态发育受到与细胞生长和分裂、及细胞过程相关的基因的调控。本研究成功构建了航天诱变向日葵lpm的SSH文库,研究结果将进一步研究lpm形成机制提供理论基础。In this paper, sunflower lpm(Long petal mutant) induced by Space Flight was used as the research material, and the SSH library of young flower buds was successfully constructed by SSH(suppression subtractive hybridization). 399 positive clones were screened from the SSH library and sequenced, and 309 Unique EST(expression sequence tags) were obtained finally. From similarity analysis in the GenBank, 121 EST were obtained with high similarites to known sequence, in which 39.16% was unique EST. By function analyzing, the genes were found related with growth, cell division and cell process, probably participating in petal morphological development of sunflower. In this study, the SSH library of sunflower lpm induced by Space Flight were contrusted successfully, and the results would provide theoretical basis to study further formation mechanism of lpm.
