摘  要:天然橡胶是产胶植物中合成的聚异戊二烯高分子化合物,而小橡胶粒子蛋白是橡胶粒子膜上参与天然橡胶分子链延伸的关键因子,与橡胶的产量和质量密切相关。本研究利用PCR技术从橡胶草中克隆得到TkSRPP5基因全长序列并进行生物信息学分析。结果表明,该基因含有一个长度为654 bp的开放阅读框,并编码217个氨基酸,所编码蛋白的分子量为23.31 kD,理论等电点为4.58。TkSRPP5为亲水性蛋白,具有24个磷酸化位点,亚细胞定位预测在细胞质中。该蛋白属于SRPP家族,二级结构主要存在α-螺旋和无规则卷曲结构,与预测的蛋白质三级结构一致。系统进化树分析表明,该蛋白与菊苣的SRPP蛋白有着最近的亲缘关系。本研究为进一步了解TkSRPP5基因的功能和其在橡胶草胶乳合成中的作用机制提供了理论依据。N atural rubber is a synthesis of isoprene polymer compounds in rubber-producing plants, and small rubber particle protein is a key factor involved in the elongation of natural rubber molecular chains on rubber particle membranes, which is closely related to the yield and quality of rubber. In this study, the full length sequence of TkSRPP5 gene was obtained from Taraxacum kok-saghyz by PCR technology and bioinformatics analysis was carried out. The results showed that TkSRPP5 contained an open reading frame with a length of 654 bp and encoded a protein of 217 amino acids with a predicted molecular weight of 23.31 kD, and the theoretical point was 4.58. TkSRPP5 was a hydrophilic protein with 24 phosphorylation sites and subcellular localization was predicted in the cytoplasm. The protein belonged to the SRPP family, and the secondary structure was mainlyα-helix and random coil, which was consistent with the predicted tertiary structure. The Phylogenetic tree analysis showed that this protein displayed the closest evolutionary relationship with the SRPP protein from Cichorium intybus. This study provides a theoretical basis for further understanding of the function of the TkSRPP5 gene and its mechanism in the synthesis of rubber grass latex.
