摘 要:为加快欧洲报春育种进程,本研究以‘庆典’、‘丹诺雅’、‘丹诺雅XL’、‘丹妮莎’、‘普利罗曼’以及上海源怡种苗股份有限公司自主育种的‘春晖’系列共38个欧报春品种为材料,利用24对SSR引物通过毛细管电泳检测法进行了欧洲报春遗传多样性和遗传关系分析。供试的24对SSR引物中11对引物稳定性和多态性较好,可用于遗传多样性和聚类分析。多态性信息指数为0.360 3~0.953 3,平均值为0.814 9。38个欧洲报春品种分别来源于6个不同品系,6个品系遗传距离都维持在较高水平,其中‘丹妮莎’和‘普利罗曼’间的遗传距离最小,上海源怡种苗股份有限公司自主育种的‘春晖’系列与‘庆典’的遗传距离最大,且‘春晖’系列与其余5个品系的遗传距离均大于0.5。通过STRUCTURE分析发现6个群体中存在5个基因库,且不同品系、不同花色的品种在基因库组成比例存在较大差异。本研究初步通过已有SSR标记开展欧洲报春遗传多样性分析,筛选出的标记可用于不同品系遗传距离的分析,但在不同品种DNA指纹图谱构建中还存在一定欠缺。后续将通过多种途径开展欧洲报春SSR标记的开发,以推进欧洲报春的育种进程以及新品种鉴定工作。In order to speed up the breeding process of European primrose, a total of 38 European primrose varieties of ’Pageant’, ’Danova’, ’Danova XL’, ’Danessa’, ’Plyroman’, and ’Chunhui’ strains which bred by Shanghai Yuanyi Seedling Co., Ltd. were used as material. 24 pairs of SSR primers were used to analyze the genetic diversity and genetic relationship of 38 European primrose varieties by capillary electrophoresis. There were 11 pairs of SSR primers with good stability and polymorphism were used for genetic diversity analysis and cluster analysis.The polymorphism information index was 0.360 3~0.953 3, with an average of 0.814 9. 38 European primrose varieties were derived from 6 different lines, and their genetic distances were maintained at a high level. Among them, the genetic distance between ’Danessa’ and ’Plyman’ is the smallest, the genetic distance between ’Chunhui’and the ’Pageant’ is the largest, and the genetic distance between ’Chunhui’ and the other five lines is greater than 0.5.Through STRUCTURE analysis, the existence of 5 gene pools in 6 populations was found, and there were significant differences in the composition ratio of gene banks in different strains and different flower colors. This study initially carried out the genetic diversity analysis of European primrose through the existing SSR markers. The selected markers can be used for the analysis of genetic distance of different strains, but there are still some defects in the construction of DNA fingerprints of different varieties. In the follow-up, the development of the European primrose SSR mark will be carried out in various ways to promote the breeding process of European primrose and the identification of new varieties.
