摘  要:目的观察木豆叶总黄酮/α-半水硫酸钙(α-calcium sulfate hemihydrate,CaS)复合材料在大鼠颅骨临界性骨缺损模型中的修复效果,积累基础实验数据。方法50只SD大鼠进行颅骨临界性骨缺损造模,随机分为5组,植入不同材料:空白对照组(无植入)、CaS组(无木豆叶总黄酮)、低剂量复合材料组(木豆叶总黄酮与CaS质量比=20 mg∶1000 mg)、中剂量复合材料组(木豆叶总黄酮与CaS质量比=40 mg:1000 mg)及高剂量复合材料组(木豆叶总黄酮与CaS质量比=80 mg∶1000 mg),每组10只。动物实验后4周、8周、12周进行评价,包括苏木素-伊红(HE)染色、Masson染色及Micro-CT定性及定量评价。结果Micro-CT三维重建影像、HE染色及Masson染色均证实空白对照组大鼠颅骨骨缺损虽有部分组织新生,但其中骨组织含量极少,骨质无法修复,说明造模成功,其余四组骨缺损均得到了修复,但修复质量CaS组较差,高剂量复合材料组最佳。骨矿物质密度(bone mineral density,BMD)和骨体积/组织体积(bone volume/total volume,BV/TV)趋势一致,在术后12周时,CaS组、低剂量复合材料组、中剂量复合材料组及高剂量复合材料组均显著优于空白对照组(P<0.05),而中、高剂量复合材料组明显优于CaS组和低剂量复合材料组(P<0.05)。结论中、高剂量木豆叶总黄酮/α-半水硫酸钙复合材料的促成骨作用明显优于单纯的α-半水硫酸钙,可以缩短骨修复时间,是一种良好的新型骨替代材料。Objective To evaluate the effect of a novel biomaterial total flavonoids of Pigeonpea(TFP)/α-calcium sulfate hemihydrate(CaS)in repairing critical cranial defects in rats.Methods Fifty SD rats were used to establish rat modes of critical cranial defect.The rat models were randomly divided into 5 groups according to different scaffold materials:the control group(no implantation),the CaS group(only CaS without TFP),the low dose composited material group(TFP∶CaS=20 mg∶1000 mg),the medium dose composited material group(TFP∶CaS=40 mg∶1000 mg)and the high dose composited material group(TFP∶CaS=80 mg∶1000 mg),with 10 rats in each.HE and Masson staining,qualitative and quantitative evaluation by Micro-CT were evaluated 4 weeks,8 weeks and 12 weeks postoperatively.Results Although there was some new born tissue in cranial defect of the control group observed by histological examination(HE and Masson staining)and Micro-CT imaging,there was little new born bone.The bone defects were not repaired,indicating that rat modes of critical cranial defect were successful.Despite bone repairs in cranial defects were all observed,the new bone quality was the best in high dose composited material group.Conversely,the CaS group was the worst.All four material groups were superior to the control group in 12 weeks postoperatively(P<0.05).Furthermore,BMD and BV/TV in the medium and high dose composited material groups were significantly higher than those in the CaS group and the low dose composited material group(P<0.05).Conclusion Medium and high dose composited material are superior toα-calcium sulfate hemihydrate,which could shorten bone repair time.
