摘  要:目的探讨染料木黄酮对氧化应激和缺血/再灌注(I/R)损伤诱导的心肌细胞凋亡的保护作用及其机制。方法采用过氧化氢诱导小鼠心肌细胞H9c2建立氧化应激模型,将细胞分为对照组、过氧化氢组和染料木黄酮组(研究组),采用TUNEL法检测细胞凋亡情况,采用免疫印迹法检测各组细胞中Bax、Bcl-2和CXCR4蛋白表达情况。通过结扎C57小鼠左前降支冠状动脉建立I/R损伤模型,将动物分为对照组、I/R损伤组(I/R组)和研究组,采用TUNEL法检测各组小鼠心肌细胞的凋亡情况,采用免疫印迹法检测各组小鼠心肌细胞中Bax、Bcl-2和CXCR4蛋白的表达情况。结果动物和细胞实验TUNEL检测结果均显示,与过氧化氢组或I/R组相比,研究组细胞凋亡数明显减少(F细胞=326.53,F动物=121.74,P<0.05);免疫印迹法检测结果显示,与过氧化氢组相比,研究组细胞中CXCR4蛋白的表达量明显降低(F=23.98,P<0.05),Bax蛋白表达量明显降低(F=14.32,P<0.05),Bcl-2蛋白表达量明显升高(F=144.76,P<0.05);研究组小鼠心肌细胞中Bcl-2/Bax比值较I/R组明显增高(F=54.16,P<0.05)。结论染料木黄酮可有效减轻心肌细胞凋亡,其可能通过降低心肌细胞CXCR4蛋白的表达而实现。Objective To investigate the protective effect of genistein against cardiomyocyte apoptosis induced by oxidative stress and ischemia/reperfusion(I/R)injury and its mechanism.Methods Mouse H9c2 cardiomyocytes were induced by hydrogen peroxide to establish a model of oxidative stress,and then the cells were divided into control group,hydrogen peroxide group,and genistein group(research group);the TUNEL method was used to measure cell apoptosis and Western blotting was used to measure the protein expression of Bax,Bcl-2,and CXCR4.C57 mice were treated with ligation of the left anterior descen-ding coronary artery to establish a model of I/R injury,and then the mice were divided into control group,I/R injury group,and research group;the TUNEL method was used to measure the apoptosis of cardiomyocytes and Western blotting was used to mea-sure the protein expression of Bax,Bcl-2,and CXCR4 in cardiomyocytes.Results The TUNEL results of both animal and cell experiments showed that compared with the hydrogen peroxide group or I/R injury group,the research group had a significant reduction in the number of apoptotic cells(Fcell=326.53,Fanimal=121.74,P<0.05);the results of Western blotting showed that compared with the hydrogen peroxide group,the research group had significant reductions in the protein expression of CXCR4(F=23.98,P<0.05)and Bax(F=14.32,P<0.05)and a significant increase in the protein expression of Bcl-2(F=144.76,P<0.05);compared with the I/R group,the research group had a significantly higher Bcl-2/Bax ratio in cardiomyocytes(F=54.16,P<0.05).Conclusion Genistein can effectively reduce cardiomyocyte apoptosis,possibly by reducing the protein expression of CXCR4 in cardiomyocytes.
