摘  要:目的研究过表达dnaJ基因对鲍曼不动杆菌生物膜形成的影响。方法以ATCC19606基因组DNA为模板扩增dnaJ基因并与质粒pWH1266双酶切后连接,筛选重组质粒并转化菌株ATCC19606,从而构建dnaJ基因过表达菌株19606-p-dnaJ,类似的办法构建空质粒对照组19606-p,用半定量结晶紫染色法观察生物膜形成的差异。结果dnaJ基因过表达组dnaJ基因相对表达量显著高于空质粒对照组,dnaJ基因过表达组24h生物膜量显著低于空质粒对照组(P<0.001)。结论过表达dnaJ基因对鲍曼不动杆菌生物膜形成能力有显著抑制作用。Objective To investigate the effect of overexpressed dnaJ on biofilm-forming ability of Acinetobacter baumannii.Methods The dna J gene was amplified from ATCC19606 genome and cloned into plasmid pWH1266.The recombinant plasmid was then transformed into strain ATCC19606 to generate the recombinant strain 19606-p-dnaJ,in which dnaJ gene was overexpressed.At the same time,the control strain 19606-p bearing empty plasmid pWH1266 was constructed by a similar method.The difference of biofilm-forming ability was then observed through the crystal violet experiment.Results The dnaJ expression was significantly higher in recombinant strain 19606-p-dnaJ than that in control strain 19606-p.However,the amount of biofilm in recombinant strain 19606-p-dnaJ was significantly lower than that in control strain 19606-p(P<0.001).Conclusion The dnaJ overexpression can significantly inhibit the biofilm-forming ability of Acinetobacter baumannii.
