提高白及种子室外萌发质量的方法研究

摘　　要：在名贵药材白及的直播育苗过程中,因为种子的自然萌发率极低,现有技术存在操作繁琐、成本投入较高、使用冷僻药品等系列问题,迫切需要利于简易推广的技术优化研究。本文针对白及种子萌发所需的四个关键要素,选取常规材料进行组合研究,通过比较白及种子吸涨膨大、出芽变色、原球茎发育及叶芽发育比例等关键参数,筛选出最优化的组合管理为:种子于4℃条件下,使用萘乙酸0.05mg·L-1+赤霉素0.01mg·L-1+0.01%蔗糖+0.01%多菌灵的1/4MS培养溶液预处理2周,然后在播种一月内每周喷雾一次0.05μg·L-1萘乙酸、0.01μg·L-1赤霉素、0.01%磷酸二胺、0.01%蔗糖、0.01%多菌灵溶液,一月后每周喷雾一次0.01%磷酸二胺、0.01%蔗糖、0.01%多菌灵溶液,这种管理方式下白及种子在2 月左右即可达到75.62%左右的出叶芽率,且无染菌现象。本研究结果利于简易推广,为白及种子直播育苗提供了技术参考。Objective:In the process of direct seedling cultivation of valuable medicinal material Baiji,because the natural germination rate of seeds is extremely low,the existing technology has a series of problems such as cumbersome operations,high cost investment,and use of cold medicines,technical optimization research that facilitates simple promotion is urgently needed.Method:This paper aimed at the four key elements required for Baiji seed germination,and selected conventional materials for combination research.By comparing key parameters such as Baiji seed imbibition and swelling,budding and discoloration,protocorm development and leaf bud development ratio,the optimal combination was selected.Result:The combination was as follows:the seeds were pretreated at 4 ℃ with naphthaleneacetic acid 0.05 mg·L-1combined with gibberellin 0.01mg·L-1 and 0.01% sucrose and 0.01% carbendazim in 1/4 MS culture solution for 2 weeks.Then the seed was planted and sprayed once a week with 0.05μg·L-1 naphthylacetic acid,0.01μg·L-1gibberellin,0.01% phosphate diamine,0.01% sucrose,0.01% carbendazim solution during one month.And the seed was sprayed once a month with 0.01% diamine phosphate,0.01% sucrose and 0.01% carbendazim solution.Under this management method,Baiji seeds can reach a leaf bud rate of about 75.62% in February without bacteria infection.The results of this study are conducive to simple promotion and provide a technical reference for Baiji seed direct seedling cultivation.
