摘  要:为了挖掘生防细菌与化学农药混用对致病疫霉的抑菌潜力,本研究将短小芽孢杆菌W-7菌株与甲霜灵锰锌混合培养后涂布统计菌落数,明确两者复配的可行性及适宜浓度。以平板对峙法检测复配液对致病疫霉菌丝体形态及生长的影响,混合培养法镜检观察复配液对孢子囊形态及直接萌发的影响,块茎切片法评价复配液在马铃薯离体组织上的防病效果。结果显示,15μg/mL的甲霜灵锰锌药液对W-7菌株的生长无明显影响,将药液与W-7菌液原液以1∶2体积比复配后,对致病疫霉菌丝体的抑制率(98.94%)及致畸率(48.56%)均明显高于两者单独处理之和;复配液处理后孢子囊的直接萌发率(5.87%)显著低于两者单独处理,同时孢子囊的畸变率(53.88%)与两者单独处理的畸变率之和相近;复配液在马铃薯块茎切片上对晚疫病的防效(91.36%)显著高于两者单独处理后的防效之和。这表明短小芽孢杆菌与甲霜灵锰锌复配在防治马铃薯晚疫病过程中,对于减少使用甲霜灵锰锌有积极作用。In order to explore the inhibition potential of mixed use of chemical pesticides and biocontrol bacterium on Phytophthora infestans,the mixture of Bacillus pumilus W-7 strain and the metalaxyl mancozeb(MM)were cultured for 48 h,and spreading plate for calculation colonies were conducted to determine the feasibility and optimum concentration of W-7 and MM compound.Dual culture method was used for detecting the effect of compound liquid(CL)on the shape and growth of P.infestans mycelia;mixture cultivation method was used for observing the effect of the mixture on shape and direct germination of pathogen sporangia;tuber slice method was used for evaluating the effect of the mixture on disease prevention.The results showed that 15μg/mL MM had no effect on growth of W-7 strain.The inhibition rate(98.94%)and the deformation rate(48.56%)of the mixture of MM and W-7 strain at 1:2 on P.infestans mycelia growth were evidently higher than that of the sum effect of MM and W-7 treatment alone.The pathogen sporangia direct germination rate treated with CL(5.87%)was remarkably lower than that of W-7 strain and MM treatment alone,and sporangia deformation rate(53.88%)after treatment with CL was much similar to that of the sum effect of MM and W-7 treatment alone.In addition,the disease prevention effect of CL(91.36%)was significantly higher than the sum effect of MM and W-7 treatment alone.These findings indicate that the mixture of W-7 strain and the metalaxyl mancozeb would greatly reduce the use of chemical pesticides for controlling potato late blight.
