摘  要:目的研究洋金花提取物对咪喹莫特诱导的银屑病小鼠模型的干预治疗作用,并对其可能的作用机制进行研究。方法每日使用咪喹莫特乳膏诱导小鼠背部皮肤产生银屑病样皮损,同时灌胃洋金花提取物(Daturae Flos extract,DFE)。每天用银屑病皮肤损伤程度评分标准(PASI评分)对小鼠背部皮肤的损伤程度进行评分。取小鼠皮肤背部组织,HE染色后观察,测量皮肤厚度;酶联免疫吸附法检测皮肤组织中肿瘤坏死因子α(TNF-α)、白细胞介素8(IL-8)的含量;用免疫组化法检测小鼠皮肤中白细胞介素17(IL^-17)、白细胞介素23(IL-23)、核转录因子κB(NF-κB)蛋白的表达水平,检测小鼠皮肤中分化簇4(CD4)、髓过氧化物酶(MPO)的表达水平来评价CD4^+T淋巴细胞、中性粒细胞在小鼠皮肤中的浸润情况。结果洋金花提取物能改善咪喹莫特诱导的小鼠背部银屑病样皮损;降低模型小鼠表皮厚度(P<0.05);降低皮肤组织中TNF-α、IL-8的表达水平(P<0.05);抑制皮肤组织中CD4+T淋巴细胞、中性粒细胞的聚集(P<0.05,P<0.01);抑制皮肤组织中IL^-17、IL-23、NF-κB的表达(P<0.05)。结论洋金花提取物对咪喹模特诱导的银屑病小鼠模型有治疗作用,其治疗作用可能是通过干预IL-23/IL^-17炎症轴和抑制NF-κB信号通路来实现。Objective To study the interventional therapeutic effect of Daturae Flos extract on imiquimod-induced psoriasis mouse model,and to explore its possible mechanism.Methods Imiquimod cream was applied daily on the back skin of mice to induced psoriasis-like lesions,and Daturae flos extract(DFE)were administered to mice.The degree of skin damage on the back of the mice was scored daily using the PASI score(PASI score)scale.The skin tissue of mice was taken for observation after HE staining,and the epidermis thickness was measured.Enzymelinked immunosorbent assay was adopted to determine tumor necrosis factor-α(TNF-α)and interleukin-8(IL-8)in mouse skin tissue;immunohistochemical assay was adopted to determine interleukin-17(IL^-17),interleukin-23(IL-23),nuclear transcription factor-κB(NF-κB)protein expression levels,and to determine cluster of differentiation-4(CD4)and myeloperoxidase(MPO)protein expression levels for evaluation of the infiltration of CD4^+T lymphocytes and neutrophils in mice skin.Results The Daturae Flos extract can ameliorate the psoriatic-like lesions induced by imiquimod on the back of mice;reduce the thickness of epidermis in mice model(P<0.05);reduce the expression levels of TNF-αand IL-8 in skin tissues(P<0.05);inhibit the infiltration of CD4+T lymphocytes and neutrophils in skin tissues(P<0.05,P<0.01);and inhibit the expression of IL-23,IL^-17 and NF-κB in skin tissues(P<0.05).Conclusion Daturae Flos extract has therapeutic effects on imiquimod-induced psoriasis of mice,and the therapeutic effects may be achieved by interfering with the IL-23/IL^-17 inflammation axis and inhibiting NF-κB signaling pathway.
