摘 要:目的基于网络药理学方法探讨西黄丸抗癌的潜在作用机制。方法经BATMAN数据库获取西黄丸的潜在活性成分及作用靶点。经DisGeNET数据库与OMIM数据库检索获取癌症疾病的相关靶点。使用Drugbank数据库获取癌症疾病的潜在治疗靶点。采用R软件对药物和疾病靶点进行匹配映射。使用String数据库构建关键靶点蛋白质互作网络,使用DAVID数据库对关键靶点行生物信息学注释,并使用R软件及Cytoscape软件进行绘图可视化。结果通过筛选得到西黄丸44个主要活性成分,共有1908个作用靶点,其中与癌症疾病靶点有关的活性成分为19个,其关键靶点涉及的相关癌种包括前列腺癌、胰腺癌、子宫内膜癌等。西黄丸可通过作用于ESR1、EGFR、SRC等靶蛋白,调节雌激素信号通路、催乳激素信号通路及ErbB信号通路等信号通路而发挥抗癌的治疗作用。结论西黄丸对癌症的治疗潜在治疗作用体现了其多成分、多靶点、多途径的药理学特点。调节癌细胞分化、抑制癌细胞增殖及血管生成、靶向雌激素受体及通路可能是其抗癌治疗的重要药效基础。Objective To investigate the potential molecular mechanism of Xihuang pill for anti-cancer treatment based on network pharmacology.Methods Potential active components and targets of Xihuang pill were obtained from Bioinformatics Analysis Tool for Molecular Mechanism of TCM database.Related targets of cancer were screened and retrieved by DisGeNET and Online Mendelian Inheritance in Man(OMIM)database.Potential therapeutic targets for anti-cancer were screened and retrieved by DrugBank.Targets of drugs and disease were matched with R program.The protein interaction network was constructed using the String database.Biological functions,metabolic pathways and the types of cancer involving key targets of Xihuang pill were analyzed by using the DAVID database.Meanwhile,R program and Cytoscape software were applied to visualize the figures.Results A total of 44 active components involved with 1908 targets were screened.Nineteen key compounds for anti-cancer were identified,which involved the related types of cancer included prostate cancer,pancreatic cancer,and endometrial cancer.Xihuang pill exerts the anticancer effects by acting on the targets including ESR1,EGFR,and SRC,and regulating the estrogen signaling pathway,the prolactin signaling pathway,and ErbB signaling pathway,etc.Conclusion Therapeutic effect of Xihuang pill on cancer is based on the its pharmacological characteristics involving multi-components,multi-targets,and regulating multi-pathways.Regulating cancer cell differentiation,inhibiting cancer cell proliferation and angiogenesis,and targeting estrogen receptors and pathways might be the important basis of its effect for treating cancer.
