摘 要:目的探讨益气补阴方对气阴两虚证糖尿病大鼠的降血糖作用及其相关机制。方法将60只SD大鼠随机分为正常组、气阴两虚模型组、糖脉康组(1.35 g·kg-1)以及益气补阴方低、中、高剂量组(1.48、2.95、5.91g·kg-1)。采用高脂高糖饲料喂养结合腹腔注射链脲佐菌素(STZ,30 mg·kg-1)建立2型糖尿病大鼠模型,在此基础上继续灌胃耗气伤阴方水煎剂(青皮∶枳壳∶附子=6∶6∶5,1.3 g·mL-1)建立气阴两虚证糖尿病大鼠模型。每天灌胃给药1次,连续8周。观察大鼠一般状态、代谢情况(24 h饮食量、饮水量及尿量)及体质量变化,每2周测定大鼠空腹血糖(FBG);给药8周后检测血清总胆固醇(TC)、甘油三酯(TG)、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)、胰岛素(Ins)、游离三碘甲腺原氨酸(FT3)、血栓素B2(TXB2)含量;采用HE染色法观察大鼠肾脏、胰腺组织病理学变化。结果与正常组比较,气阴两虚模型组大鼠的代谢量(24 h饮食量、饮水量及尿量)以及血清FBG、TC、TG、MDA、Ins、TXB2含量均显著升高(P<0.01),体质量、SOD、GSH、FT3含量均显著降低(P<0.01)。与气阴两虚模型组比较,益气补阴方各剂量组大鼠体质量、代谢水平、TC及MDA含量变化均不明显(P>0.05);益气补阴方高、中剂量组大鼠的FBG、TG、TXB2含量显著下降(P<0.01),GSH、FT3含量显著升高(P<0.05,P<0.01);益气补阴方高剂量组大鼠的SOD含量显著升高(P<0.05),Ins含量显著下降(P<0.05),肾脏、胰腺组织病理损伤明显减轻。结论益气补阴方能有效改善气阴两虚证糖尿病大鼠的糖脂代谢紊乱,其作用机制可能与提高机体抗氧化能力,修复肾脏、胰腺病理损伤,减轻胰岛素抵抗,保护心血管功能有关。Objective To explore the hypoglycemic effect and mechanism of Yiqi Buyin formula on diabetic rats with Qi and Yin deficiency.Methods Sixty SD rats were randomly divided into normal group,Qi and Yin deficiency model group,low,medium and high dose groups(1.48,2.95,5.91 g·kg-1)of Yiqi Buyin formula,and Tangmaikang group.Type 2 diabetic rat models were established by giving high fat and sugar feed combined with intraperitoneal injection of streptozotocin(STZ,30 mg·kg-1);diabetic rat models with Qi and Yin deficiency were established by gavage of excessive Citri reticulatate pericarpium viride,Aurantii fructus and Aconiti lateralis radix praeparata(6∶6∶5,1.3 g·mL-1)on this basis.Rats were given intragastric administration once a day for 8 weeks.The general state,metabolism(24 hours food and water intake,and urine volume)and body weight change of rats were observed.The fasting blood glucose(FBG)of rats was measured every 2 weeks.The contents of total cholesterol(TC),triglyceride(TG),superoxide dismutase(SOD),malondialdehyde(MDA),glutathione(GSH),insulin(Ins),free triiodothyronine(FT3)and thromboxane B2(TXB2)were measured,pathological changes of kidney and pancreatic tissue in rats were observed by HE staining after 8 weeks of administration.Results Compared with the normal group,the contents of metabolism,FBG,TC,TG,MDA,Ins,TXB2 increased significantly(P<0.01),the body weight and the contents of SOD,GSH,FT3 of rats decreased significantly(P<0.01)in Qi and Yin deficiency model group.Compared with Qi and Yin deficiency model group,the body weight,metabolic levels and the content of MDA and TC were not significantly different(P>0.05)in rats of all dose groups of Yiqi Buyin formula;the contents of FBG,TG,TXB2 decreased significantly(P<0.01),the contents of GSH,FT3 increased significantly(P<0.05,P<0.01)in rats of the high,medium dose groups of Yiqi Buyin formula;the content of SOD increased significantly(P<0.05),the content of Ins decreased significantly(P<0.05),the pathological damage of the kidney and pancreas tissu
