摘 要:目的比较不同代数人头皮毛乳头(Dermal papilla,DP)细胞体外培养方法及其形态学差异。方法从人头皮分离DP细胞进行原代培养,传代培养至第9代,观察、记录不同代数细胞形态、多层聚集性变化。结果1~3代DP细胞呈梭形,具有明显的多层聚集生长特性;而4代以上形状不规则DP细胞逐渐增多,细胞传代时间从低代的2~3周延长至6周以上,多层聚集性逐渐消失。结论体外培养的DP细胞随着代数次数增加,其生长速度、形态、多层聚集性等特性发生显著改变。Objective To compare the morphological difference among different generations of human scalp dermal papilla cells cultured in vitro.Methods Primary human scalp dermal papilla cells were isolated and cultured in vitro to the9 th generation,and the morphological characteristics and multilayer aggregation of each generation were observed and recorded.Results Dermal papilla cells of the 1 st-3 rd generations were spindle-shaped with obvious multilayer aggregation growth while those of the 4 th and younger generations were increasingly irregular-shaped with multilayer aggregation gradually disappearing.Besides,passage time extended from two to three weeks at the older generations to more than six weeks at the younger generations.Conclusion Such features as growth rate,morphology and multilayer aggregation of dermal papilla cells cultured in vitro,change significantly as passage proceeds.
