摘  要:颗粒细胞是卵巢中必需的体细胞,在卵泡发育过程中起重要作用。为探究miR-10b对牛卵巢颗粒细胞凋亡的影响,本实验中2个处理组分别转染miR-10b模拟物(miR-10b mimics)和miRNA模拟物(mimics NC),转染48h后,通过Quantitativereal-timePCR(qRT-PCR)检测内源miR-10b表达量,明确miR-10b mimics转染成功。利用qRT-PCR和Western Blot检测凋亡相关蛋白Caspase-3、Bcl-2和Bax表达量的变化;利用流式细胞术检测颗粒细胞凋亡率。结果显示:与mimics NC组相比,miR-10b mimics组Bcl-2 mRNA和蛋白表达水平显著降低,Bax和Caspase-3 mRNA和蛋白表达水平显著升高;同时miR-10b mimics组的细胞凋亡率较mimics NC组显著升高。结果表明,miR-10b可促进牛卵巢颗粒细胞凋亡。Granulosa cell is an essential somatic cell in ovary and plays an important role in follicular development.The present study was performed to investigate the effects of miR-10b on apoptosis of bovine ovarian granulosa cell.Bovine ovarian granulosa cell was transfected with miR-10b mimics and mimics NC,respectively.After transfection for 48 h,the expression of endogenous miR-10b was detected by Quantitative real-time PCR(qRT-PCR)to confirm the successful transfection of miR-10b mimics.The expression levels of apoptosis-related proteins Caspase-3,Bcl-2 and Bax were detected by qRT-PCR and Western Blot;and granulosa cell apoptosis rate was detected by flow cytometry.The results showed that forced expression miR-10b significantly decreased Bcl-2 expression at both the mRNA and protein levels(P<0.05),while the mRNA and protein levels of Bax and caspase-3 were significantly increased(P<0.05)in granulosa cells.Furthermore,the apoptosis rate in miR-10b mimics group was significantly higher than that in mimics NC group(P<0.05).These results indicated that miR-10b could promote the apoptosis of bovine ovarian granulosa cells.
