摘 要:目的建立同时测定长柱金丝桃中金丝桃苷和槲皮素含量的高效液相色谱分析方法。方法采用高效液相色谱法,色谱柱为Zorbax SB-C18柱(5μm,250 mm×4.6 mm),流动相为甲醇∶乙腈∶0.01 mol/L磷酸(10∶30∶60),流速1.0 m L/min,检测波长357 nm,柱温35℃。结果金丝桃苷和槲皮素分别在10.12~50.6μg和8.2~41μg范围内,进样量与色谱峰面积呈良好线性关系,r值分别为0.999 2和0.999 1;平均回收率(n=6)分别为99.13%和100.8%,RSD分别为2.26%和2.35%。结论本实验方法法操作简便、准确、稳定性及重复性好,可用于同时对长柱金丝桃中金丝桃苷和槲皮素成分的含量测定。Objective To establish HPLC method for the determination of hyperoside and quercetin in Hypericum ascyron L.. Methods The Zorbax SB-C18(5μ m,250 mm ×4.6 mm) column was adopted and eluted with mobile phase consisted of methanol: acetonitrile:0.01 mol/L aqueous phosphoic acid(10: 30: 60), at the column temperature 35℃. The flow rate was 1.0 mL/min and detective wave length was 357 nm. Results Hyperoside was proved to be linear in the range of10.12 ~50.6μ g with a regression coefficient of 0.9992,the average recovery was 99.13%, with RSD of 2.26%. Quercetin was proved to be linear in the range of 8.2 ~41μ g with a regression coefficient of 0.9991, the average recovery was 100.8%, with RSD of 2.35%. Conclusion The method is simple, accurate, stable and repeatable and can be used to determine the content of hyperoside and quercetin in Hypericum ascyron L.
