摘 要:目的:探讨左归丸含药血清对高糖负荷下体外培养ICR小鼠早期胚胎影响的蛋白组学机制。方法:以ICR小鼠早期胚胎为研究对象,分别用普通胚胎培养液、40mmol/L葡萄糖的胚胎培养液、40mmol/L葡萄糖加5%左归丸含药血清的胚胎培养液进行体外培养,通过iTRAQ方法分析得到培养液中差异蛋白并分析。结果:与空白组比较,有34个差异蛋白在高浓度葡萄糖作用后显著下调,经左归丸含药血清干预后回调,这些蛋白通路主要集中在核糖体、丙酮酸代谢、糖酵解/糖异生、雌激素信号转导途径、抗生素合成等通路。高糖组和左归丸组间涉及差异蛋白通路除上述外,还与PI3K-Akt信号、EB病毒感染、抗原处理和呈现等通路相关。结论:高浓度葡萄糖不利于胚胎发育与健康,左归丸含药血清可通过调节培养液中相关蛋白通路保护胚胎发育及改善或预防糖耐量低减、糖尿病等相关疾病,也可为中医“肾主发育”理论提供进一步的实验依据。Objective:To investigate the proteomic mechanism of Zuogui Pills containing serum on ICR mouse embryos cultured in vitro under high glucose load.Methods:The early embryos of ICR mice were taken as the research object,and cultured in vitro with normal embryo culture medium,40mmol/L glucose,40mmol/L glucose and 5%Zuogui Pills drug serum.The different proteins in the culture medium were obtained and analyzed by iTRAQ method.Results:Compared with the blank group,there were 34 differentially expressed proteins that were significantly down-regulated after glucose action,and then reactivated after intervention by Zuogui Pills containing serum.These protein pathways mainly focused on ribosomes,pyruvate metabolism,glycolysis/gluconeogenesis,estrogen signal transduction and antibiotic synthesis pathways.In addition,it is also related to the PI3K-Akt signaling,EB virus infection,antigen processing and presentation pathway.Conclusion:High concentration of glucose is not conducive to embryonic development and health.Zuogui Pills serum can protect embryonic development and improve or prevent impaired glucose tolerance,diabetes and other related diseases by regulating the related protein pathway in the culture medium.And this can provide a certain experimental basis for the theory of‘kidney governing growth and development’.
