摘 要:目的:观察新风胶囊对佐剂关节炎(AA)大鼠ETS-1、β-catenin及血小板活化的影响。方法:将48只大鼠随机分为正常对照组、模型对照组、来氟米特组、新风胶囊组,每组12只。除正常对照组外,其余大鼠复制成AA模型。第19天开始给药,各组连续用药30d。观察肺功能变化,ELISA检测血清细胞因子的表达,流式细胞仪检测外周血GMP-140、CD40L表达,Western blotting检测肺组织ETS-1、β-catenin、E-cadherin、GMP-140、PAF蛋白。结果:与正常对照组比较,模型对照组肺功能参数FEV1、FEF50、FEF75、PEF显著降低(P<0.01),细胞因子TGF-β1、IL-17、IL-21表达显著升高(P<0.01),IL-10显著降低(P<0.01),外周血GMP-140、CD40L显著升高(P<0.01),ETS-1表达显著降低(P<0.01),β-catenin、E-cadherin及GMP-140、PAF表达显著升高(P<0.01,P<0.05);与模型对照组比较,新风胶囊组肺功能参数FEV1、FEF50、FEF75、PEF均显著升高(P<0.05,P<0.01),细胞因子TGF-β1、IL-17、IL-21表达显著降低(P<0.05,P<0.01),IL-10表达显著升高(P<0.01),外周血GMP-140、CD40L降低(P<0.01,P<0.05),ETS-1显著升高(P<0.01),β-catenin、E-cadherin及GMP-140、PAF表达显著降低(P<0.05)。结论:新风胶囊通过调控ETS-1表达,抑制β-catenin激活,降低血小板活化,抑制免疫炎症反应,改善类风湿关节炎肺功能。Objective:To observe the effects of Xinfeng Capsules on ETS-1,β-catenin and platelet activation in adjuvant arthritis(AA)rats.Methods:A total of 48 rats were randomly divided into normal control(NC)group,model control(MC)group,leflunomide(LEF)group and Xinfeng Capsules(XFC)group,12 rats in each group.Except for the NC group,the remaining rats were replicated into the AA model.Dosing was started on the 19th day,and each group was administered continuously for 30 days.The changes of pulmonary function were observed.The expression of serum cytokines were detected by ELISA.The expression of GMP-140 and CD40L in peripheral blood were detected by flow cytometry.ETS-1,β-catenin,E-cadherin,GMP-140 and PAF proteins were detected by Western blotting.Results:Compared with the NC group,the pulmonary function parameters(FEV1,FEF50,FEF75,PEF)of the MC group were significantly reduced(P<0.01),the expressions of cytokines TGF-β1,IL-17,IL-21 were significantly increased(P<0.01),IL-10 was significantly decreased(P<0.01),GMP-140 and CD40L in peripheral blood were significantly increased(P<0.01),the expression of ETS-1 was significantly decreased(P<0.01),β-catenin,E-cadherin,GMP-140 and PAF were significantly increased(P<0.01,P<0.05).Compared with the MC group,the pulmonary function parameters(FEV1,FEF50,FEF75,PEF)of the XFC group were significantly higher than those of the MC group(P<0.05,P<0.01),the expressions of cytokines TGF-β1,IL-17 and IL-21 were significantly decreased(P<0.05,P<0.01),and the expression of IL-10 was significantly increased(P<0.01),GMP-140 and CD40L in peripheral blood were significantly decreased(P<0.01,P<0.05),ETS-1 was significantly increased(P<0.01),β-catenin,E-cadherin,GMP-140 and PAF were significantly decreased(P<0.05).Conclusion:Xinfeng Capsules can inhibit the activation ofβ-catenin,inhibit platelet activation,inhibit immune inflammatory response and improve RA pulmonary function by regulating ETS-1 expression.
