摘 要:目的:优选复方三黄脂质体中黄芩、大黄提取物的大孔树脂纯化工艺。方法:以有效部位黄芩总黄酮、大黄游离蒽醌,有效成分黄芩苷、大黄素和大黄酚的吸附洗脱率为指标,采用单因素试验考察树脂类型、上样速度、上样浓度、洗脱剂种类与用量等对复方三黄脂质体中黄芩、大黄提取物大孔树脂纯化工艺的影响。结果:最优纯化工艺为:选定AB-8大孔树脂,上样浓度为每毫升含生药0.2g,最大上样量为4BV,上样速度为4BV/h,用5BV的水以4BV/h的速度洗脱,弃去洗脱液,再用12BV的80%乙醇以4BV/h的流速洗脱,收集洗脱液。结论:优选的纯化工艺合理稳定,有效成分转移率高,适于生产应用。Objective:To optimize the purification technology of extracts from Sastellaria baicalensis and Rhizoma Rhei in Compound Sanhuang liposome with macroporous resin.Methods:Taking adsorption-desorption rate of effective fractions and active ingredients as indexes,the effect of the type of macroporous resins,the concentration of sample solution a,type and dosage of elution agent on purification technology of extracts from Sastellaria baicalensis and Rhizoma Rhei in Compound Sanhuang liposome were investigated by single factor test.Results:The optimal purification procedure was using AB-8 macroporous resin.The content of sample solution was 0.2g/mL.the volume was 4BV;the flow rate was 4BV/h.To elute with distilled water,the volume was 5BV and the flow rate was 4BV/h,giving up the eluate.To elute with 80%ethanol,the volume was 12BV and the flow was 4BV/h gathering the elute.Conclusion:This optimized purification technology was reasonable and stable,the amount of active component was high.The technology could be extended to large-scale production application.
