摘 要:目的:探讨生脉散对糖尿病大鼠心脏收缩功能的保护作用。方法:高脂饲料喂养大鼠4周后腹腔注射无菌链脲佐菌素(STZ)45mg/kg,血糖值≥16.7mmol/L即为造模成功。成模大鼠随机分为模型组、曲美他嗪组、生脉散组。检测大鼠糖耐量、超声心功能、在体血流动力学、血脂4项、心肌横截面积、心肌过氧化物酶体增殖物激活受体(PPARα)蛋白表达。结果:糖尿病大鼠心脏功能及结构均发生显著改变,与对照组比较,模型组心脏重量指数(HMI)、左心室舒张末压(LVEDP)显著增高(P<0.01,P<0.05),左心室收缩期压力最大变化速率(Vdp/dt_(Max))、左心室射血分数(EF)、心肌组织PPARα表达降低(P<0.05,P<0.01)。与模型组比较,两给药组HMI、LVEDP显著降低(P<0.05),Vdp/dt_(Max)、EF、PPARα表达均显著增高(P<0.05,P<0.01),两给药组间比较无显著性差异。结论:生脉散可抑制糖尿病所致的心肌肥厚,并保护糖尿病大鼠心脏收缩功能,但不参与调节动物血糖、血脂和胰岛素抵抗。其对糖尿病大鼠心功能的影响可能与PPARα有关。Objective:To explore the protective effect of Shengmai Powder(SMP)on cardiac contractile function in diabetic rats.Methods:Rats were intraperitoneally injected 45 mg/kg streptozotocin(STZ)after had been fed 4 weeks high-fat diet.The rats blood glucose≥16.7 mmol/Lwere considered as successfully induced to diabetic model.The diabetic rats were randomly divided into model group,trimetazidine(TMZ)group and SMP group.Glucose tolerance,echocardiographic cardiac function,in vivo cardiac hemodynamics,four items of blood lipid,cross-sectional area of myocardium,expression of PPARαprotein were investigated.Results:Cardiac function and structure of diabetic rats were significantly changed.Compared with the control group,the cardiac weight index(HMI),and the left ventricular end diastolic pressure(LVEDP)of the model group were significantly increased(P<0.01,P<0.05),the maximum change rate of left ventricular systolic pressure(dp/dtMax),the left ventricular ejection fraction(EF)and the expression of PPARαdecreased(P<0.05,P<0.01).Compared with the model group,the HMI and LVEDP of the TMZ group and SMP group were significantly decreased(P<0.05),the dp/dtMax,EF and PPARαexpression significantly increased(P<0.05,P<0.01).There was no difference between TMZ group and SMP group.Conclusion:Shengmai Powder can inhibit the cardiac hypertrophy induced by diabetes and protect the contractile function in diabetic rats,but it is not involved in the regulation of blood glucose,blood lipid and insulin resistance.The effects of PPARαon the cardiac function of diabetic rats may be related to PPARα.
