育麟方对卵巢储备功能减退大鼠卵巢颗粒细胞超微结构及功能的影响

摘　　要：目的:通过分组干预环磷酰胺所致的卵巢储备功能减退(DOR)模型大鼠,研究育麟方对卵巢颗粒细胞(GCs)超微结构的影响及可能机制。方法:腹腔注射环磷酰胺制备DOR成熟雌性SD大鼠模型,分成空白组,模型组,中药低、中、高剂量组,补佳乐组,分别灌胃21d,予动情前期处死,测定大鼠血清抑制素B(INHB)表达水平,计算卵巢指数,电镜观察各组GC超微结构。结果:与模型组比较,中药低、中剂量组及补佳乐组卵巢指数显著增高(P<0.05)。与模型组比较,中药低、中、高剂量组及补佳乐组血清INHB显著增高(P<0.05)。电镜下中药低剂量组GC胞体较大,糖原颗粒较多,具有成簇脂滴,线粒体形态正常,数量较多。结论:育麟方对DOR大鼠GC的超微结构具有改善作用,可改善颗粒细胞功能,促进INHB分泌,抑制卵泡凋亡,改善卵巢内卵母细胞的数量和质量,从而治疗DOR。Objective:To study the effects of Yulin Formula on the ultra structure of ovarian granulosa cells(GC)in diminished ovarian reserve(DOR)and its possible mechanism by intervening the DOR model rats which were caused by cyclophosphamide(CTX).Methods:The mature female SD rat model of DOR was made by intraperitoneal injection of CTX.They were randomly divided into five groups:control group,model group,low,medium and high dose group of traditional Chinese medicine,progynova group.The rats were administrated by gavage for 21 d respectively,and were killed before estrus.The INHB expression level in serum was measured,the ovarian index was calculated,and the ultra structure of ovarian granulosa cells in each group was observed by electron microscope.Results:Compared with the model group,the ovarian index of the low and medium dose groups and progynova group was significantly increased(P<0.05).Compared with the model group,the INHB expression level of serum in low,medium and high dose group of traditional Chinese medicine,progynova group was significantly higher(P<0.05).Under electron microscope,compared with the model group,the GC cell bodies in the Yulin Formula low dose group were larger under electron microscope,with more glycogen granules,and there were a large number of clustered distribution of intracytoplasmic lipid droplets,mitochondria were normal in morphology and large in number.Conclusion:Yulin Formula has an improvement effect on the ultra structure of GC in DOR rats.It can improve the function of granulosa cells,promote the secretion of IHNB,inhibit the apoptosis of follicles,and improve the quantity and quality of oocytes in the ovary,so as to treat the DOR.
