摘  要:目的:应用16SrRNA技术,探讨在醋酸刺激下,干姜大枣提取物对大鼠肠道微生态及血清胃泌素(GAS)、血管活性肠肽(VIP)的影响。方法:将30只SD大鼠随机分为对照组、醋酸组和醋酸药物组,每组10只。对照组正常饮食,其余两组采用pH值为3.9、温度为4℃的醋酸灌胃。醋酸药物组大鼠灌胃给予干姜大枣提取物1mL/100g收集大鼠粪便,分析评价干姜大枣提取物对实验大鼠肠道菌群属水平结构及多样性的影响。结果:与对照组比较,醋酸组在肠道菌群属水平的多样性改变方面较大(P<0.05),出现了9种存在差异的菌属。与醋酸组比较,醋酸药物组出现了一些新的菌群改变情况。乳酸菌属在醋酸组中的相对丰度较低,梭菌属、肠球菌属在醋酸组中的相对丰度呈现上升趋势。与对照组比较,醋酸组GAS、VIP的含量显著升高(P<0.05);与醋酸组比较,醋酸药物组GAS、VIP的含量显著降低(P<0.05)。结论:干姜大枣提取物能够有效改善醋酸引起的肠道菌群失调,促进有益菌群的生长、抑制致病菌及条件致病菌的繁殖。Objective:To investigate the effects of 16SrRNA on intestinal microecology,serum gastrin(GAS)and vasoactive intestinal peptide(VIP)in rats stimulated by acetic acid.Methods:Thirty SD rats were randomly divided into control group,acetic acid group and acetic acid drug group.Taking pH concentration 3.9,the temperature of 4℃fixed acetic acid,to fill the stomach.The rats in acetic acid group were given Zingiberis Rhizoma and Jujubae Fructus extract 1 mL/100 g.Before sampling,fecal samples of each rat were directly collected to analyze and evaluate the effect of Zingiberis Rhizoma and Jujubae Fructus extract on the structure and diversity of the intestinal flora of the experimental rats at the level of genus.Results:The diversity of intestinal flora in the acetic acid group was significantly higher than that in the control group(P<0.05),and there were 9 different species of bacteria in the acetic acid group.Compared with the acetic acid group,the acetic acid group showed some new changes in microflora.The relative abundance of lactobacillus in acetic acid group is low,and that of clostridium and enterococcus shows an upward trend.GAS and VIP contents in acetic acid group were significantly higher than those in the control group(P<0.05).GAS and VIP levels in acetic acid group decreased compared with acetic acid model group(P<0.05).Conclusion:It was found that the Zingiberis Rhizoma and Jujubae Fructus extract can effectively improve intestinal flora disorder caused by acetic acid,promote the growth of beneficial flora,inhibit the reproduction of pathogenic bacteria and opportunistic pathogens.
