摘 要:目的:观察消癌解毒方对肝癌大鼠脂质代谢的影响。方法:76只雄性SD大鼠按正常组、模型组、消癌解毒方低剂量组和高剂量组随机平均分组,予模型组、低剂量组和高剂量组饮用0.01%二乙基亚硝胺(DEN)溶液建立肝癌模型。第5周开始分别予低剂量组和高剂量组10.87和21.74g·kg^-1·d^-1的消癌解毒方灌胃。第16周处死所有大鼠,取肝脏,TMT肽段标记后进行质谱鉴定,分析差异蛋白并用Western blotting验证结果。结果:由KEGG分类可知差异蛋白主要富集于代谢类别,主要与脂质代谢相关。对通路层面的信息进行富集分析,可见在类固醇激素生物合成(Cyp2c11、Hsd3b5、Cyp2e1等)、脂肪酸降解(Acox1、Acsl1、Adh6等)、PPAR信号通路(Gk、Slc27a5、Lpl等)通路上有明显富集,标志性蛋白的表达差异经Western blotting验证后得到证实。结论:消癌解毒方可能通过干预肝癌大鼠的脂质代谢来抑制肿瘤的发生发展。Objective:To investigate the effect of Xiaoai Jiedu Decoction on lipid metabolism in DEN-induced hepatocellular carcinoma rats.Methods:A total of 76 SD male rats were randomly divided into normal group,model group,lowdose group and high-dose group.The model group,the low-dose group and the high-dose group were given 0.01%DEN solution for free drinking to establish the hepatocellular carcinoma model.The low-dose group and the high-dose group were given the dose of 10.87 and 21.74 g·kg^-1·d^-1 respectively to gavage the hepatocellular carcinoma model from week 5.At week 16,liver tissue samples were taken for TMT peptide labeling and mass spectrometry identification,analyze differential proteins,and detect the expression levels by using Western blotting.Results:It could be seen from the classification of KEGG that the difference proteins were mainly concentrated in the metabolic categories,which were mainly related to lipid metabolism.Enrichment analysis of the information of metabolic differences related proteins on pathways,the biosynthesis of steroid hormones(Cyp2c11,Hsd3b5,Cyp2e1,etc.),fatty acid degradation(Acox1,Acsl1,Adh6,etc.),PPAR signaling pathways(Gk,Slc27a5,Lpl,etc.)had the obvious enrichment.The results of the analysis of the iconic protein expression differences were confirmed by Western blotting.Conclusion:Xiaoai Jiedu Decoction may inhibit the development of tumor by intervening the lipid metabolism process of Hepatocellular Carcinoma rats.
