摘  要:目的:对臭牡丹降糖有效部位化学成分进行研究。方法:臭牡丹乙醇提取物分别用石油醚、乙酸乙酯、正丁醇萃取得到3个提取部位;利用体外α-葡萄糖苷酶活性测定3个提取物的α-葡萄糖苷酶抑制活性来初步筛选臭牡丹降糖有效部位,然后通过高效液相-四级杆-飞行时间-质谱(HPLC-Q-TOF-MS/MS)研究降糖有效部位的化学成分,并通过查阅相关文献初步确定哪些化学成分具有潜在的α-葡萄糖苷酶抑制活性。结果:通过体外α-葡萄糖苷酶活性测定确定乙酸乙酯部位为臭牡丹降糖有效部位,并从该部位鉴定出29个化合物,包括12个苯乙醇苷,7个黄酮,5个酚酸以及5个其它化合物;通过查阅相关文献,初步确定化合物2~4、7、9~10、12~13、15、19、26具有潜在的α-葡萄糖苷酶抑制活性。结论:利用体外测定α-葡萄糖苷酶活性结合HPLC-Q-TOF-MS/MS技术能快速筛选臭牡丹降糖有效部位以及研究其化学成分;最终,明确了臭牡丹体外降糖的主要化学成分,为寻找更好的α-葡萄糖苷酶抑制剂提供理论依据。Objective:To determine the active chemical constituents of hypoglycemic effective parts from Clerodendron bungei Steud.Methods:Tested fractions were obtained by partitioning the ethanol extracts of Clerodendron bungei Steud using petroleumether,ethyl acetate and butyl alcohol.The active hyperglycemic parts were screened byα-glycosidase enzyme activity in vitro,then the chemical constituents of the effective fraction of hypoglycemic substances were studied by high performance liquid phase quadrupole time of flight mass spectrometry(HPLC-Q-TOF-MS/MS),and the related literature was used to determine which chemical components have potentialα-glucosidase inhibitory activity.Results:Ethyl acetate and part exposed proper hypoglycemic activity through the determination ofα-glucosidase activity in vitro,and 29 compounds were identified from the part,including 12 phenylethanoid glycosides,7 flavonoids,5 phenolic acids and 5 other compounds.It was preliminarily determined that the compounds 2-4,7,9-10,12-13,15,19,26 have potentialα-glucosidase inhibitory activity by consulting the relevant literature.Conclusion:The effective fractions of the hypoglycemic parts from Clerodendron bungei Steud and its chemical constituents can be rapidly screened by the determination ofα-glucosidase activity in vitro combined with HPLC-Q-TOF-MS/MS technology.Finally,the main chemical components of the hypoglycemic parts from Clerodendron bungei Steud in vitro are determined,providing a theoretical basis for finding betterα-glucosidase inhibitors.
