摘 要:目的观察高压氧(HBO)联合还原型谷胱甘肽(GSH)对大鼠下肢缺血再灌注损伤(IRI)的影响.方法将40只大鼠随机分为5组(每组8只):假手术组(S组)、缺血再灌注组(IR组)、高压氧组(HBO组)、还原型谷胱甘肽组(GSH组)、高压氧+还原型谷胱甘肽组(HBO+GSH组).通过夹闭腹主动脉,建立大鼠急性下肢IRI模型.再灌注后,测定并比较各组间大鼠血清的炎症因子[白细胞介素-8(IL-8)和肿瘤坏死因子α(TNF-α)]和氧化应激指标[超氧化酶岐化酶(SOD)和血清丙二醛(MDA)]水平.结果与IR组比较,HBO组和GSH组大鼠的血清IL-8、TNF-α及MDA水平更低,SOD水平更高;与HBO组、GSH组比较,GSH+HBO组血清IL-8、TNF-α及MDA水平降低更明显,SOD水平升高更加明显(均P<0.05).结论HBO和GSH均可减轻下肢IRI引起的炎症反应和氧化应激水平,其中二者联合使用时效果更加明显.Objective To observe the effect of HBO combined with reduced glutathione(GSH)on lower limb ischeinia-reperfiision injury(IRI)in rats.Method Forty rats were randomly divided into Five groups(8 in each group):sham operation group(S group),ischemia repcrfiision group(IR group).hyperbaric oxygen group(HBO group),reduced glutathione group(GSI 1 group),and reduced glutathione group(HBO+GSH group).Rat acute lower limb IRI model was established by rLamping abdominal aorta.After reperfusion,serum levels of inflammatory factors[interleukin-8(IL-8)and tumor necrosis factor(TNF-α),and oxidative stress indicators superoxidase dismutase(SOD)and scrum malondialdehyde(MDA)].were measured and compared.Results Compared with the I/R group,the serum IL-8,TNF-αand MDA levels of the rats in the F1BO group and the GSF1 group were lower and the SODlevels were higher.Compared with the F1BO group and the GSF1 group,the scrum IL-8,TNF-αand MDA levels in the GSH+HBO group decreased more significantly,while the level of SOD increased more significantly(all P<0.05)Conclusion Both HBO and GSH can reduce the level of inflammatory response and oxidative stress caused by lower limb IRI,and the effect is more obvious when they arc used together.
