摘 要:基因组注释预测了AX-3普遍存在于不同植物种类,但其功能鲜有报道。本研究通过StAX-3基因反义cDNA在本氏烟(Nicotiana benthamiana)体内表达,实现了其AX-3基因表达下调;蛋白质组表达谱测定分析显示,伴随AX-3积累减少,168个DAPs(表达倍绝对值大于1.5)的积累随之改变;其中15个密切关联DAPs网络分析显示,AX-3下调表达,主要降低了9种线粒体内代谢酶类的积累,包括氧化磷酸化ATP合成酶和TCA循环核心脱氢酶,同时,增加了光合作用代谢酶类合成,包括光合磷酸化ATP合成酶和叶绿素合成底物萜类酶等。叶绿素和ATP含量等表型测定结果,证实了萜类底物和叶绿素合成积累增加,促进光合磷酸化ATP合成积累。结果表明,StAX-3调控氧化和光合磷酸化ATP合成积累模式。该结果不仅明确了St AX-3具有调控线粒体和叶绿体能量代谢的功能,而且为进一步研究植物ataxin-3功能提供理论指导。Ataxin-3 is annotated and predicted to be present in widely plant genomes.Little is known on the function of plant ataxin-3.In this study,an antisense St AX-3 cDNA from potato was expressed in N.benthamiana.Proteomic profile in ataxin-3 down-regulated Nicotiana benthamiana transgenic lines were assayed and analyzed.Result showed that expression of 168 DAPs,larger than 1.5 on expression-fold was significantly changed along with reduction on StAX-3 expression.Analysis on a STRING network composed of 15 DAPs showed that 9 mitochondrial components,including core dehydrogenases in TCA cycle and ATP biosynthases in the oxidative phosphorylation were reduced along StAX-3 down-expression.Meanwhile,biosynthesis on terpenoid backbone as the substrate for chlorophyll,and ATP in photo phosphorylation were intensified while St AX-3 expression was down-regulated.Assay result on chlorophyll and ATP contents confirmed that ATP accumulation promoted by photo phosphorylation was resulted from enhancing chlorophyll biosynthesis,due to increase on terpenoid substrate.The results indicate that St AX-3 regulates ATP accumulation model of oxidative and photo phosphorylation.The result not only specifies StAX-3 function on energy metabolism regulation between mitochondrion and chloroplast,also sets a basis for further study on StAX-3 function.
