摘 要:目的建立高效液相色谱法(HPLC)测定地耳草中槲皮苷和异槲皮苷含量的不确定度评定方法。方法采用高效液相色谱(HPLC)外标法测定地耳草中槲皮苷和异槲皮苷的含量并建立数学模型,分析测量不确定度的影响因素。结果HPLC外标法测定地耳草中槲皮苷和异槲皮苷的含量分别为0.40%和0.39%。置信概率为95%时,槲皮苷与异槲皮苷的扩展测量不确定度分别为5.06×10^-4和9.12×10^-4,含量测定结果分别为(0.40±0.05)%和(0.39±0.09)%(k=2)。结论本文建立的不确定度评定方法,可用于HPLC法测定地耳草中槲皮苷和异槲皮苷含量测定的不确定度分析,对地耳草的质量控制有指导意义。Objective To establish a method of uncertainty analysis for content determination of quercitrin and isoquercitrin in Hypericum japonicum Thunb.ex Murray by high performance liquid chromatography(HPLC).Methods HPLC external standard method was conducted for content determination of quercitrin and isoquercitrin in Hypericum japonicum Thunb.ex Murray,and the mathematical model was established to systematically analyze and evaluate the influential factors of measurement uncertainty.Results The content of quercitrin and isoquercitrin in Hypericum japonicum Thunb.ex Murray was 0.40%and 0.39%by HPLC external standard method,respectively.When the confidence probability was 95%,the expanded uncertainty was 5.06×10^-4 and 9.12×10^-4(k=2),respectively,and the content was(0.40±0.05)%and(0.39±0.09)%(k=2),respectively.Conclusion The evaluation method of the measurement uncertainty can be used to determine the uncertainty of quercetin and isoquercitrin in Hypericum japonicum Thunb.ex Murray,and has a practical significance for quality control of Hypericum japonicum Thunb.ex Murray.
