摘  要:目的:研究海洛因滥用者尿液中代谢物吗啡与06_单乙酰吗啡丙酰化的时间、温度条件,为GC/MS法检测吗啡与06-单乙酰吗啡提供最优检测条件。方法:空白尿液中添加吗啡与O^6-单乙酰吗啡,分别在温度60℃、70℃ 80℃.90℃ 95℃水浴加热,进行10min、20min、30min、40min、50min、60min丙酰化反应,GC/MS测定吗啡、单乙酰吗啡丙酰化产物的含量。结果:丙酰化温度709C、时间40min时尿液中吗啡、O^6-单乙酰吗啡丙酰化反应较完全,GC/MS检测效果更好。结论:GC/MS法检测海洛因吸食者尿液中代谢物吗啡与06-单乙酰吗啡时,丙酰化温度控制在70、时间为40min时获得较好丙酰化反应效果,检测更加准确。Objective To study the time and temperature conditions in propionylation reaction of Morphine and O*-monoacetylmorphine in the urine of heroin abusers and to provide the optimal detection conditions for method of GC/MS of morphine and O^6-monoacetylmorphine.Method Morphine and 0°-monoacetylmorphine were added to the blank urine,the propionylation reaction was at a temperature of 60℃、70℃ 80℃.90℃ 95℃ and the time of 10mins,20mins,30mins,40mins,50mins and 60mins under heating in water bath.The content of propionylation reaction products of morphine and O^6-monoacetylmorphine was determined by GC-MS.Results The propionylation reaction of morphine and 06-monoacetylmorphinein in the urine was complete at the temperature 70℃ and the time 40mins.And can get better GC/MS determination of the effect.Conclusion GC/MS method was used to detect the morphine and O*-monoacetylmorphine in urine of heroin abusers.The propionylation temperature was controlled at 70℃ and the time was 40mins,the better propionylation reaction was obtained,and the detection was more accurate.
