摘  要:目的:观察霉茶蛋白对自发性高血压大鼠(SHR)肾脏的保护作用,并借助肾小管上皮细胞(NRK-52E)对其作用机制进行探讨。方法:将40只雄性SHR随机分为4组:模型组、复方罗布麻片组(阳性药物,50 mg·kg^-1)、霉茶蛋白低、高(70,140 mg·kg^-1)剂量组,每组10只。复方罗布麻片组和霉茶蛋白组早晚各灌胃给药1次,容量为10 ml·kg^-1,连续给药7周,模型组给予等体积蒸馏水。末次给药后1 h处死动物,迅速剥离肾脏组织,HE染色观察肾脏组织结构形态变化;测定肾脏组织Toll样受体4(TLR4)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)mRNA表达;血管紧张素Ⅱ(AngⅡ)刺激NRK-52E后,经霉茶蛋白各剂量(0.05,0.1,0.2,0.4μg·ml^-1)干预24 h,收集细胞,测定各组TLR4、IL-6、TNF-αmRNA表达。结果:霉茶蛋白可改善SHR肾脏组织的炎性病变,降低SHR肾脏组织和NRK-52E细胞中TLR4、IL-6、TNF-αmRNA表达水平,与模型组比较,差异有统计学意义(P<0.05)。结论:霉茶蛋白对SHR肾脏具有一定的保护作用,其机制可能与下调炎性因子TLR4、IL-6、TNF-α的表达有关。Objective:To observe the protective effect of Meicha protein on kidney of spontaneously hypertensive rats(SHR),and to explore its mechanism of action in combination with renal tubular epithelial cell NRK-52 E.Methods:Totally 40 male SHRs were randomly divided into 4 groups:the model group,compound apocynum tablets group(positive drug,50 mg·kg^-1),Meicha protein low-dose and high-dose groups(70 and 140 mg·kg^-1)with 10 ones in each group.SHRs were given the relevant medicine intragastrically,and the model group was given constant volume of distilled water intragastrically in the morning and afternoon everyday for7 weeks.Renal tissue was quickly excised after the last administration,the histomorphological changes of kidney were observed by hematoxylin-eosin(HE)staining;mRNA expressions of Toll-like receptor 4(TLR4),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in renal tissues were determined by RT-PCR.AngiotensinⅡstimulated NRK-52 E was treated with various doses of Meicha protein(0.05,0.1,0.2 and 0.4μg·ml^-1)for 24 h,and the cells were collected for real-time PCR analysis of mRNA expressions of TLR4,IL-6 and TNF-αin each group.Results:Meicha protein can improve the inflammatory lesions of SHRs’kidney tissue and decrease the expressions of TLR4,IL-6 and TNF-αmRNA in SHRs’kidney tissue and NRK-52 E cells,and when compared with the model group,the differences were statistically significant(P<0.05).Conclusion:Meicha protein has a certain protective effect on SHRs’kidney,and its mechanism may be related to the down-regulation of mRNA expressions of inflammatory factors TLR4,IL-6 and TNF-α.
