摘 要:目的:考察水凝胶型和橡胶型壮骨麝香止痛膏中挥发性成分体外透皮差异。方法:该研究采用垂直式Franz扩散池,以离体乳猪腹部皮肤为渗透屏障进行体外透皮扩散试验。使用20%聚乙二醇400-生理盐水溶液作为接收液,转速300 r·min^-1,样品透过液处理方法为2414.9×g离心5 min,残留样品处理方法为加无水乙醇20 ml、超声40 min。以GC法测定樟脑、薄荷脑、冰片、水杨酸甲酯的累计渗透量、膏片残留量、皮肤滞留量,比较水凝胶型和橡胶型壮骨麝香止痛膏中挥发性药物的体外透皮特性。结果:两种工艺制备的壮骨麝香止痛膏体外透皮特性基本一致,水凝胶型工艺制备的壮骨麝香止痛膏,24 h上述药物累积透过量高于橡胶型工艺制备的壮骨麝香止痛膏;水凝胶型壮骨麝香止痛膏中上述药物在皮肤中的滞留量低于橡胶型;药物经皮渗透后,上述药物在水凝胶膏片中的残留量高于橡胶型。结论:该研究初步证明水凝胶型壮麝香止痛膏中挥发性药物的体外经皮渗透行为与橡胶型基本一致,但是挥发性药物的透皮能力有所加强,具有渗透速率快、渗透量高的特点。Objective:To study the difference of in-vitro transdermal permeation of volatile components from hydrogel-type and rubber-type of Zhuanggu Shexiang Zhitong Gao.Methods:The modified vertical Franz diffusion cells were used to conduct a transdermal experiment in vitro with the isolated abdominal skin of Suckling pigs as the transdermal absorption barrier.20%Polyethylene glycol400-saline solution was used as the receiving solution at a stirring speed of 300 r·min^-1.The sample was centrifugated at a speed of2414.9×g for 5 min.The residual sample was treated with 20 ml of absolute ethanol and 40 min of ultrasound.The cumulative permeability,sample residue and skin retention of camphor,menthol,borneol and methyl salicylate were determined by GC.At the same time,the in-vitro transdermal permeability of volatile drugs from hydrogel type and rubber type of Zhuanggu Shexiang Zhitong Gao was compared.Results:The in-vitro transdermal properties of the two formulations of Zhuanggu Shexiang Zhitong Gao were basically the same.The cumulative drug permeability of hydrogel type in 24 hours was higher than that of rubber type,and the retention of volatile drugs in skin of hydrogel type was lower than that of rubber type.In addition,the residue of volatile drugs in hydrogel type was higher than that in rubber type.Conclusion:The experiment preliminarily proved that the in-vitro percutaneous permeation behavior of volatile drugs from hydrogel-type Zhuanggu Shexiang Zhitong Gao is basically the same as that of rubber type.However,the transdermal ability of volatile drugs is enhanced by hydrogel,and hydrogel products have the characteristics of rapid penetration rate and large penetration amount.
