摘  要:利用分光光度法以对硝基月桂酸酯为底物溶液对诺维信脂肪酶435进行活性表征。结果表明:诺维信脂肪酶435作用的最适温度为40~60℃,最适pH为8.00~11.00,Ba^2+、K^+,Tween-20,EDTA和PMSF都可以适当提高酶的活性,而去污剂Triton X-100以及Cu^2+、Ca^2+、Zn^2+则会使酶活性有不同程度的降低。另外反复使用10次后诺维信脂肪酶的活性仍然在80%左右。该研究可以为该酶的在生物柴油制备等应用提供一定的数据参考。The activity of Novozymes lipase 435 was characterized by spectrophotometry with p-nitrolaurate as substrate solution.The results showed that the optimum temperature of Novozymes lipase 435 was 40-60℃,the optimum pH was 8.00-11.00.Ba^2+,K^+,Tween-20,EDTA and PMSF all could properly increase the activity of the enzyme,while the detergent Triton X-100 and Cu^2+,Ca^2+,Zn^2+ could reduce the activity of the enzyme to varying degrees.In addition,the activity of Novozymes lipase 435 was still about 80%after repeated use for 10 times.This study can provide some data reference for the application of the enzyme in biodiesel preparation and so on.
