摘 要:目的探究白癜风患者外周血中亚油酸(linoleic acid,LA)的水平及其对外周血CD8^+T细胞的作用。方法利用超高效液相色谱-质谱非靶向代谢联用技术检测8例白癜风患者及8例健康对照者血清中代谢物LA的水平,采用流式细胞术检测5例白癜风患者及5例健康对照者外周血中CD8^+T细胞的活化及杀伤性分子表达情况,最后体外常规培养5例患者外周血CD8^+T细胞,予以LA刺激,明确LA对白癜风患者外周血CD8^+T细胞活化及杀伤分子表达的影响。结果与健康对照相比,白癜风患者血清中LA水平显著升高(P<0.05),且外周血CD8^+T细胞过度活化(P<0.05)并高表达毒性杀伤分子颗粒酶B(granzyme B,GzmB,P<0.05)、穿孔素(perforin,PRF,P<0.05);进一步体外试验发现LA显著抑制白癜风患者外周血CD8^+T细胞的活化及毒性杀伤效应分子的表达。结论进展期白癜风患者血清中LA水平上调,可抑制CD8^+T细胞活化及杀伤效应功能,帮助机体重建自身免疫紊乱后平衡状态。Objective Explore the role of linoleic acid(LA)from the peripheral blood of vitiligo patients on CD8^+T cells.Methods The serum of 8 vitiligo patients and 8 healthy controls were detected by the high-performance liquid chromatography-Q-TOF method.The activation and effector function of CD8^+T cells in peripheral blood of 5 vitiligo patients and 5 healthy controls were measured by flow cytometry.Then,the effects of high linoleic acid(LA)on the activation and effector function of CD8^+T cells in peripheral blood of 5 vitiligo patients were tested in vitro.Results The serum levels of LA in vitiligo patients were significantly higher than those in healthy controls(P<0.05).We observed the overactivation of CD8^+T cells and the release of granzyme B and perforin in vitiligo patients(P<0.05),and we found that LA could significantly suppress the activation and killing function of CD8^+T cells in peripheral blood of vitiligo patients.Conclusion Serum LA upregulate in patients with advanced vitiligo.LA can inhibit the activation and killing function of CD8^+T cells in vitro and may involve in helping the body rebuild its autoimmune homeostasis.
